Objects To determine the expression of COX-2,P53,and Caspase-3 in the tissue of chronic cholecystitis,gallbladder adenomatous polyp and gallbladder cancer,and investigate the correlation between their expression level,biological behavior and pathological feture in gallbladder cancer.Methods The expression of COX-2,P53,and Caspase-3 in the tissue of 30 patients with chronic cholecystitis and 19 with gallbladder adenomatous polyp and 25 with gallbladder cancer were examined by immunohistochemistry.The statistical analysis was accomplished by SPSS12.0 software using x~2 test.Results The rate of positive expression of COX-2 in primary gallbladder cancer, gallbladder adenomatous polyp and chronic cholecystitis were 76%(19/25),42.1%(8/19)and 23.3%(7/30)respectively,those of P53 were 68%(17/25),31.6%(6/19)and 6.7%(2/30),those of Caspase-3 were 32%(8/25), 42.1%(8/19)and 76.7%(23/30).The expression of COX-2 in late Nevin stage(Ⅳ-Ⅴ) cases was higher than that in early Nevin stage(Ⅰ-Ⅲ)(P<0.05)and significant difference was shown between cases with and without metastasis(P<0.05).Positive correlation was found between P53 and COX-2 in gallbladder adenomatous polyp~a and gallbladder cancer~b tissue,(P~a=0.001,P~b=0.038),negative correlation was found between COX-2 and Caspase-3(P~a=0.025,P~b=0.001).Conclusions The expression of COX-2 and P53 were higher in gallbladder cancer and precancerous adenomatous polyp,whereas Caspase-3 was lower.The pattern of the expression of COX-2,P53 and Caspase-3 in gallbladder adenomatous polyp tissue indicated that they could be early events of gallbladder cancer.The mutation of P53 up-regulated the expression of COX-2,which regulated that of Caspase-3,and then the apoptosis was inhibited.That indicated the three proteins could be taken part in the progression of gallbladder cancer.Positive correlation was found between P53 and COX-2 in gallbladder adenomatous polyp and gallbladder cancer tissue,negative correlation was found between COX-2 and Caspase-3.The expression of COX-2,P53 and Caspase-3 could be related with the Nevin stage and metastasis.
