Expression And Clinical Significance Of HOXA5Protein In Gallbladder Carcinoma

Objective: We design to detect the expression of HOXA5protein in bothbenign and malignant gallbladder diseases; With the clinicopathologicaldata collectied, we also analyze the factors that may affect the expression ofHOXA5protein. Accordingly, we probe to explore the role and significance ofthe HOXA5protein in the development of benign and malignant gallbladderdiseases.Methods: We collected35cases of gallbladder carcinoma specimens aswell as30cases of gallbladder polyp specimens and20cases of chroniccholecystitis specimens treated in Department of General Surgery, TongjiHospital, Tongji Medical College, Huazhong University of Science andTechnology, from September2009to2011August, as the carcinoma group,polyp group and cholecystitis group respectively. All these cases shouldhave complete medical records informations and should not have gongthrough radiotherapy, chemothrapy or other adjvent therapy before surgery.All specimens were fixed in10%formalin, embedded in paraffin，and cut into4μm serial sections. Immunohistochemistry was used to observe theexpression of HOXA5protein in these tissues. The relationship between theexpression of HOXA5protein and the related clinicopathlogical parameterswere analysed statistically in case-control study.Result: The expression intensity of HOXA5protein in the chroniccholecystitis group and the gallbladder polyp group were significantly(P<0.005) higher than gallbladder carcinoma group, while there was nodistinct difference between the gallbladder polyp group and the chroniccholecystitis group(P=0.15); In the gallbladder polyp group, the expressionintensity of the ones greater than1.0cm were obviously (P=0.006) lowerthan the ones less than1.0cm;80%(4/5) of the negative expression cases inthe gallbladder polyp group, showed a dysplasia of the mucosal epithelium;In the gallbladder carcinoma group, the positive expression cases weremostly the ones with well-differentiated pathological type(P=0.015),whichdemonstrated few clinical evidence of lymph nodes or distantmetastasis(P=0.03).Conclusion: The high expression intensity of HOXA5protein in thechronic cholecystitis group and the gallbladder polyp group were notobviously affected by the factor such as age, gender, and gallstone. Theexpression intensity of HOXA5protein is significantly low in the gallbladdercarcinoma group, which is closely relatied with the stage，grade and clinicalprognosis of gallbladder carcinoma; The HOXA5protein may play an important role during the development from precancerous lesions togallbladder cancer, so it helps to early diagnosis even in the stage ofprecancerous lesions by detecting the expression intensity of HOXA5protein;...
