Objective:Glucocorticoid is anabolismed mainly by the adrenal cortex.It is concerned with stress and immunity antiphlogistic and so on.Glucocorticoid is applied at clinical medicine comprehensively.Part obturation is comprehensive at orthopaedics.But glucocorticoid is hard to avoid exosmic.The investigation is a few that glucocorticoid damnify musculature.It is merely report internal that glucocorticoid induce muscle atrophy.The causation is ill-defined.In this experiment,we pick Zelanian White Rabbit.To investigate morphologic effect and histological changes of gluteal muscle effected by glucocorticoid with intramuscular injection.Methods:The experiment was conducted at the laboratory of Tianjin General Hospital, Tianjin medical university between June 2007 and November.2007.Twenty-four healthy adult rabbits were selected and randomly.Assigned into two groups with twelve rabbits in each group:normal control group and test group.Each rabbit was tested as auto-control on one sidegluteal muscle in the experimental groups.Parts of gluteal muscles were sliced up and dyed with hematoxylin-eosin staining(HE).The slides were observed under light microscope and electron microscope analyzed with image analysis system.Results:A total of Twenty-four animals were involved in the result analysis.①: Histological detection results:The HE dyed slides and the tested muscle slides showed that muscle fibers arrayed orderly.Muscle cells kept integrated.No necrosis was found.Nucleus arrayed tidily in the out part of muscle cells near cytomembrane.Sarcomere were integrated.Diameter tested muscle fibers decreased obviously.Muscle cells number was increased each per unit area.Fiber size and number of the auto——control sides and all change in the course of the test,which was different to that in the normal control group.②Electron microscope detection results: Normal rabbit gluteal muscle of rabbits was mixed type muscle with typeⅠand typeⅡfibers blend up.TypeⅠfibers were smaller than typeⅡfibers.The size of typeⅡ muscle fibers of the test side muscle were found reduced.TypeⅡfibers electron density increased more significant than typeⅠfiber.TypeⅡfibers electron density were asymmetric and arranged distortion.TypeⅠfibers changed insignificant.There was insignificant difference between the fiber type at the experimental sides and the muscle fiber in the normal control group.Conclusion:Glucocorticoid induce muscle atrophy confirmly,mainly of typeⅡmuscle fibers.But it has insignificant effect on typeⅠmuscle fibers.
