ObjectiveProthymosin alpha(ProTα)is a highly initially unstructured protein,and widely exists in all mammalian tissues.ProTαplays a very important role in cell.Thymosin beta is a newly discovered special substance which can stimulate cell movement,and is highly expressed in many tumor tissues.It is reported that the higher experssion was related to latent metestasis positively.Lots of domestic and international scholars have sutdied on the expression of tumor indicators in bladder cancer,but have little study on the expression and relationship between thymosin and bladder cancer.This experiment have studied on the expression of prothymosinαand thymosin beta in bladder cancer and normal bladder tissue by the means of immunohisochemisty,and explored its significance in tumor diagnosis and treatment.Materials and Methods1.Materials and Methods76 cases of bladder cancer tissues,all from the patients underwent sugrical resection in the Second Hospital of Tianjin Medical University bewteen 1995 to 2001. 10 cases of bladder tissues,all from the patients underwent benign prostatic hyperplasia sugrical resection.These tissues are fixed by 4%paraformaldehyde, embedded by paraffin.2.Clinical and pathological files76 cases of transitional cell carcinoma of the bladder,includeing 56 male and 10 female.Accoding to WHO international tumor organization classification is classified: G124 cases,G228 cases,G324 cases;TNM:Ta-126 cases;T2-450 cases.All these cases are not have chemotherapy and radiotherapy beofer opeartion.10 cases of normal mucous membrane of urinary bladder.3.Main reagents and soucreThe primary antibody anti-human thymosin beta4,10,15,prothymosinαand CD34 were porvided by Tianjin Runtek Teconology Limited Company.4.MethodsImmunohistoehemisty:The protein expression of thymosin beta4,thymosin beta 10,thymosin beta 15 and prothymosinαin71 eases of bladder cancer tissues and 20 cases of the normal tissue were examined by the means of SABC,while PBS was used as negative controls.Results judging:Buffy in cells' is considered as positive. cases defined as positive were regarded by both staining intensity and the percentage of immunoreactive tumor cells percentage.5.Statistical analysisUsing Chi-squaer test:0.01<P<0.05,difference was significant;P≤0.01, difference was remarkably singifieant.Results1.In 76 cases bladde cancer tissues,the expression of thymosin beta4,thymosin beta10,thymosin beta 15 and prothymosinαwere buffy fine grain in bladde cancer and the normal bladder tissue,which were mainy located in tumor cells'cytoplasmic,in the state of diffusion.The experssion rate of thymosin beta4,thymosin beta10,thymosin beta 15 and prothymosinαwas 89.5%(68/76),82.9%(63/76)and 93.4%(71/76).The experssion had remakrab difference with the normal tissue.2.The expression rate of thymosin beta4,15 and prothymosinαprotein had positive correlation with the stage and grade of bladder cancer.The expression of prothym0sinαprotein had positive correlation with the stage and grade of bladder cancer recurrence.But the expression of thymosin beta4,10,15 protein had none positive correlation with the stage and grade of bladder cancer recurrence.The expression of thymosin beta4,10,15 and prothymosinαprotein had none positive correlation with the sex and age of bladder cancer.3.The distribution and quantity of microvessel had difference in the tumor tissue.There were none expression of thymosin beta10 and 15.It hint that thymosin beta10 and 15 have no effect on angiogenesis.The protein of thymosin beta 4 and prothymosinαwere found in bladder vascular endothelial cell.It hint that thymosin beta4 and prothymosinαhave effect on angiogenesis.Microvessel density of thymosin beta4 and prothymosinαprotein postive endothelial cell,had positive correlation with the stage and grade of bladder cancer.The microvessel density of thymosin beta4 and prothymosinαprotein positive tissue had none positive correlation with the sex and age of bladder cancer.Conclusion1.The expression of thymosin beta4,15 and prothymosinαprotein had positive correlation with the stage and grade of bladder cancer.2.Microvessel density had positive correlation with the stage,grade and recurrence of bladder cancer.3.The expression of prothymosinαprotein had positive correlation with the stage and grade of bladder cancer recurrence.But the expression of thymosin beta4,10,15 protein had none positive correlation with the stage and grade of bladder cancer recurrence.4.Thymosin beta4 and prothymosinαcan promote bladder cancer cells proliferation and metastasis by directly and promoring angiogenesis.
