To compare the difference of activity in vitro and vivo and toxicity between PEG thymosin α1 and thymosin α1, and to study the amelioration of pharmacodynamics, pharmacokinetics, toxicity of Tal after polyethylene glycol(PEG)modification, The pharmacodynamics and toxicities of PEG Tα1 were observed in this paper. In the pharmacodynamic tests, when compared with Tα1, the administered frequencies of PEG Tα1 were fewer, the administered period was prolonged, and further more the activities in vitro was kept, when the two factors acted together, the biological effect of drug on liver injury were improved. Results of all toxicity tests illustrated PEG Tα1 had overcome anaphylaxis of Tal which happened most frequently in all adverse reactions and had no remarkable toxicity on test animals especially on immune system. In conclusion, after PEGylation, many properties of Tal had changed, which indicated PEG Tal could solve the problems of Tα1 such as short half-life, frequent administration, high cost and so on. PEG Tα1 is expected to find a place as a valuable alternative to normal Tα1, which show improved patient convenience and compliance.
