Object:This issue observatea PEDF expression in the retina by the high oxygen-induced retinal neovascularization in mice animal models with injecting GM6001 in vitreous cavity.Methods:48 C57BL/6J mice(96 eyes)age of 7 days in SPF were randomly divided into 4 groups:normal,high-oxygen group,GM6001 intervention group and the phosphate buffer control group,each 12.The normal group mice fed in normal air environment,another three groups of rats placed in oxygen boxes,establish a high oxygen-induced retinal neovascularization animal model.GM6001 intervention group rats injected GM6001 1μl into the vitreous cavity at the 12th days after birth,and phosphate buffer control group was injected with the same volume of phosphate buffer,normal and high-oxygen group did not deal with.The mice were executed at 17th days after birth.Removed left eye making specimens,with anti-PEDF and anti-CD31 antibodies respectively for immunohistochemical biopsy pathology observed;Separateing from the right eye,extracting retinal tissue protein for analysis the content of MMPS-2,MMPS-9 and PEDF in the retina with ELISA.Result:compared to the normal group a large number of neovascular breakthrough the retinal tissue membrane High-oxygen group.Obviously the model of the the retinal neovascularization is successful.Compared to PBS treatment group there is significantly narrowed area retinal neovascular in GM6001 treatment group(P<0.01).immunohistochemistry results showed that in normal retinal tissue,PEDF strong expression in all layer of retina,especially in ganglion cell layer and nerve fiber layer.outer nuclear layer also expressed,high oxygen group and the PBS control group pedf expression weakened in all layer of retina only in the ganglion cell layer and nerve fiber layer has been significantly expression.PEDF strong expression in all layer of retina in GM6001 treatment group,the expression similar to the normal group.ELISA showed that MMP-2,MMP-9,PEDF in the normal group,GM6001 group,the control group and PBS high-oxygen groups were statistically differences in the P<0.01 GM6001 group and the normal group was not statistically different P>0.05,high-oxygen group and PBS no significant difference between the control group P>0.05.PEDF in the first 17 days of the retina of the content of PBS GM6001 group 1.7 times the control group,MMP-2 in the retina of GM6001 group PBS 0.47 times the control group,MMP-9 in the retina of the content of PBS control group GM6001 Section 0.36 times.Conclusion:injecting GM6001 1μl into the vitreous cavity with a certain dose by inhibiting MMP-2 and MMP-9 can effectively strengthen the expression of PEDF in the oxygen-induced retinopathy in mice and the growth of neovascularization was inhibited.
