Background:Allergic rhinitis(AR) is one of common diseases in Otorhinolaryngology-Head Neck surgery. It is chronic inflammatory disease in nasal mucosa, which release mediators of inflammination that mediated by IgE when atopy individual contact allergen, and many immune cell and cytokine participate. At present, drug treatment, as nasal corticosteroids and H1-receptor antagonist is one of methods in treat AR.Objective: The purpose of this study is to investigate the expression of aquaporin 5(AQP5) and the effects of Fluticasone propionate(FP) and Levocabastine(LEVO) nasal spray applied on its expression ,in our study, we first established AR rat models with ovalbumia intraperitoneall. Then AR rats were treated by FP or LEVO. Immumohistochemistry and RT-PCR methods were applied to detect the expression and the distribution of AQP5 in the level of protein and mRNA in nasal mucous membrane with AR. The study will reveal primarily the effect on AQP5 of FP and LEVO in AR.Methods:1.40 cases Wister RATS were divided randomly into AR (30 cases) and control (10cases) groups. AR models in Wister rat were induced in intraperitoneall by injecting ovalbumia (OVA) on alternate days lasting 8 times.Then AR group were instilled with OVA sustain allergic reaction. Control group injected with 0.9% Nacl instead of OVA. Observe the pathological and praxiology score when AR group appearance typical allergic rhinitis symptom.2.When allergic rhinitis models were established successfully, the AR model rats were divided randomly into allergic rhinitis group treated by fluticasone propionate (F group,10cases), allergic rhinitis group treated by levocabastine group(L group,10 cases) and untreated allergic rhinitis group (AR group,10cases), After three groups were provoked with OVA every day, F group were treated 200 mg per day(50 mg per puff,two puffs in each nostril), L group 200 mg per day(50 mg per puff,two puffs in each nostril) .On the 28th day, Immumohistochemistry and RT-PCR methods were applied to detect the expression and the distribution of AQP5 in the level of protein and mRNA in nasal mucous membrane with AR.Results :1.The expression of AQP5 in the level of protein: The distribution of AQP5 was in accordance in all groups in rat nasal mucous. The expression of AQP5 was positive in the membrane and cytoplasm of the cilia epithelium and the glands. The expression of AQP5 was significantly difference between F group and L group and AR group (p < 0.05), but the quantity of AQP5 was not significance difference between L group and AR group (p﹥0.05).The quantity of AQP5 was no significance difference between F group and control group (p﹥0.05),the expression of AQP5 was significantly difference between L group and control group(p < 0.05).2. The expression of AQP5 in the level of gene: The products sited in 740bp fragment ,the expression of AQP5 mRNA was respectively 0.73±1.04,1.10±0.10,1.02±0.20,0.79±0.18 in every group. The q-test results were consistent to the above study.Conclusions:1. High expressions of AQP5 in rat nasal mucosa with AR indicate. AQP5 might be one of pathological factors of AR that concered with gland's excessive secretion and organization's edema.2. Glucocorticoid can down regulate the expression of AQP5 and reduce gland's secretion and nasal obstruction by changing the expression of AQP5.3. H1-receptor antagonist can't reduce the expression of AQP5. That induce H1-receptor antagonist treat allergic rhinitis may be unrelated with the expression of AQP5.
