Objective To investigate the effect of fibrin glue as perivenous support on preventing intimal and medial proliferation of vein graft and to explore the effect of that on preventing vein graft stenosis.Methods Eighteen New Zealand rabbits were divided into experimental group(n=9) and control group(n=9). All animals unilateral jugular vein were anastomosed end to end to common carotid artery as vein grafting model. Fibrin glue was applied as perivenous support in experimental group at the same time, while fibrin glue was not applied in control group. The vein grafts were harvested after 10 weeks. The intimal and medial thickness of vein graft were measured by computer assisted analysis system. The proliferation of smooth muscle cell(SMC) was detected by PCNA , Ki-67 and SMA stain. The data was analysised by SAS statistic program.Results Under light microscopy, thinner intimal and medial were observed in experimental group compared with those in control group(intimal:28.11±9.8μm vs 105.47±15.75μm,P<0.0001; medial: 93.71±18.50μm vs 125.93±15.84μm, P<0.0001). Less smooth muscle cells stained by SMA in intimal were found in experimental group compared with those in control group. PCNA and Ki-67 positive cells detected by IHC were hardly found in the intimal and medial region in both group.Conclusion Fibrin glue as a perivenous support of vein graft could decrease proliferation of SMC in intimal and medial and significantly reduce the intmal and medial thickness , therefore fibrin glue could prevent vein graft stenosis.
