The expressions of Skp2 and PTEN at protein level in NSCLC tissues of 62 patients and 17 adjacent mormal tissues of lung were detected by immunohistochemical S-P method.Results:(1) The expression of Skp2 expressing in NSCLC tissues at transcription level was higher than that in adjacent normal tissues of lungs ,between of which there exists significant statistical difference(p≈0.000). PTEN mRNA of NSCLC tissues was lower than that of adjacent normal tissues of lungs and there was significant statistical differenc(ep≈0.000)of the differential expression of PTEN mRNA between NSCLC tissues and adjacent normal tissues of lungs. The expression of p27mRNA of NSCLC tissues was also lower than that of adjacent normal tissues of lungs. There also exists significant statistical difference(p=0.006)of the differential expression of p27 mRNA between NSCLC tissues and adjacent normal tissues of lungs.(2) After Pearson Chi-Square test, the expression of skp2mRNA of NSCLC tissues was inversely correlated with that of PTEN mRNA and p27 mRNA in NSCLC tissues, respectively (rs=?0.863,p<0.05; rs=?0.852,p<0.05.). There exists positive correlation between PTEN mRNA and p27 mRNA expressing in NSCLC tissues(rs=0.902,p<0.01).(3) PTEN protein of NSCLC tissues was obviously lower than that of adjacent normal tissues of lungs. There was significant difference of PTEN protein between NSCLC tissues and adjacent normal tissues of lungs(p<0.01).Skp2 protein in NSCLC tissues was markedly higher that of adjacent normal tissues of lungs. There also exists significant difference of PTEN protein between NSCLC tissues and adjacent normal tissues of lungs(p<0.01). PTEN protein and Skp2 protein were significantly correlated with differentiation, TNM staging and lymphonode metabasis of NSCLC tissues (p<0.01or p<0.05) and were not associated with gender, age and tissue types (p>0.05).Conclusion: Skp2 of NSCLC was up-regulated at both transcriptional level and protein level, while PTEN was down-regulated at both transcriptional level and protein level. p27 mRNA show down-regulated tendengcy in NSCLC tissues.
