PART ONE EFFECTS OF JIEJUFUSHENNING ON ADVANCED GLYCATION END PRODUCTS IN SERUM AND EXPRESSION OF RECEPTOR FOR ADVANCED GLYCATION END PRODUCTS IN KIDNEYObjective: To investigate the effects of Jiejufushenning(JJFSN) on advanced glycation end products(AGEs) in serum and expression of receptor for advanced glycation end products(RAGE) in kidney.Methods: Diabetic SD rats were induced by injection of streptozotocin, then the diabetic rats were randomly divided into four groups: diabetic control group (group B), Jiejufushenning group(group C), aminoguanidine group (group D), Jiejufushenning and aminoguanidine group (group E), age-matched norma1 SD rats served as norma1 control group(group A). After 12 weeks of treatment by corresponding intervention, body weight and fasting blood glucose(FBG) were determined by the normal methods. The levels of AGEs in serum were determined by fluorospectrophotometry, and the expressions of RAGE in kidney were determined by immunohistochemical methods.Results: The levels of FBG, AGEs in serum and expressions of RAGE in kidney in rats of group C, group D and group E were obviously lower than those of group B, and the levels of group D were lower than those of group C, and the levels of group E were the lowest, but still higher than those of group A(P<0.05);body weight of group B was the lowest.Conclusion: Similar to AG, JJFSN has effect on decreasing the levels of AGEs in serum and the expression of RAGE in kidney. JJFSN has also effect on decreasing FBG. Combination of JJFSN and AG have a significantly cooperated action. PART TWO EFFECTS OF JIEJUFUSHENNING ON TRANSFORMING GROWTH FACTOR-BETA 1 IN SERUM AND URINE AND PROTECTIVE EFFECTS ON THE KIDNEY OF DIABETIC RATSObjective: To investigate the effects of Jiejufushenning(JJFSN) on transforming growth factor-beta 1 in serum and urine and protective effects on the kidney of diabetic rats.Methods: Diabetic SD rats were induced by injection of streptozotocin, then the diabetic rats were randomly divided into four groups: diabetic control group (group B), Jiejufushenning group(group C), aminoguanidine group (group D), Jiejufushenning and aminoguanidine group (group E), age-matched norma1 SD rats served as norma1 control group(group A). After 12 weeks of treatment by corresponding intervention, fasting blood glucose(FBG), body weight, kidney weight, kidney weight/body weight(KW/BW), urinary volume of 24 hours, the quantitation of urine protein of 24 hours (24hPro), blood urea nitrogen(BUN), serum creatinine(Scr), urine creatinine, cholesterin(TC) and triglyceride (TG) were determined by the normal methods. the levels of transforming growth factor-beta 1(TGF-β1) in serum and urine were determined by enzyme linked immunosorbent assay(ELISA); the morphological structure of renal tissues was observed by light microscope and electron microscope.Results: The levels of FBG, KW/BW , urinary volume of 24 hours, 24hPro, BUN, Scr, TC, TG, TGF-β1 in serum and urine in rats of group C, group D and group E were obviously lower than those of group B, but higher than those of group A, and the levels of group D were lower than those of group C, and the levels of group E were the lowest(P<0.05). The levels of urine creatinine in rats of group C, group D and group E were obviously higher than that of group B, but lower than that of group A.Conclusion: JJFSN and AG have effect in protecting kidney of diabetic rats, the mechanisms might relate to decreasing the levels of TGF-β1 in serum and urine, combination of JJFSN and AG have a significantly cooperated action.
