Objective: To study the inhibitory effect of a COX-2 inhibitor celecoxib on the growth of human nasopharyngeal carcinoma CNE-2Z cell lines and potential mechanisms, and research the expression of COX-2 in CNE-2Z cells .To explore the value of celecoxib in the chemoprevention and chemotherapy against nasopharyngeal carcinoma and offer the feasibility and corresponding theory evidence for subsequent animal and clinical experiment.Methods: The effect of celecoxib on the growth of CNE-2Z cells was evaluated by MTT assay; The expressions of proliferating cell nuclear antigen (PCNA) and cyclooxygenase-2(COX-2) were assessed by immunocytochemistry method ;Cell cycle was measured by flow cytometry (FCM) after being incubated with celecoxib; The protein expression of caspase-3 was examined using western-blot.Results:(1)The data of MTT showed that celecoxib significantly suppressed the growth of CNE-2Z cells in time and dose-dependent manner. The live cells decreased gradually with the increasing of drug concentration and disposal time. (2) Celecoxib inhibited the proliferation of CNE-2Z cells.The expression of PCNA markly decreased after being incubated with celecoxib for 48 hours.(3)The expression of COX-2 was positive and it decreased with the increasing of drug concentration .(4) The cell cycle was arrested in G0/G1 phase in CNE-2Z cells treated with celecoxib .(5) Western-blot showed that celecoxib concentration was directly proportional to the caspase-3 protein expression .Conclusion:1. The expression of COX-2 in human nasopharyngeal carcinoma CNE-2Z cells was positive.2. celecoxib suppressed the growth of CNE-2Z cells by inhibiting proliferation and keeping the cell cycle in G0/G1 phase.3.The mechanisms of celecoxib inhibited the growth of CNE-2Z cells were related to COX-2, PCNA and caspase-3 may be one of important mechanisms of its anti-angiogenic effect.4.It is significant for celecoxib to be studied in the prevention and therapy of nasopharyngeal carcinoma.COX-2 inhibitors are promised to play an important role in the chemoprevention and chemotherapy against nasopharyngeal carcinoma...
