ObjectiveThis study was design to evaluate the effect and safety of Matrine on the prevention of posterior capsular opacification in rabbit eyes .Methods36 New Zealand rabbits weighing 1.8 to 2.2kg were used .The animals were randomly divided into 4 groups. The phacoemulsification was performed in the right eyes,and the drugs were injected into the anterior chamber after the operation.Three different concentrations of Matrine(0.5g/l,1.0g/l,1.5g/l) were given 0.1ml into the capsula lentis.Group A using BBS 0.1ml served as a control group . The changes of cornea ,anterior chamber were observed everyday in 2 weeks postoperatively; intraocular pressure were observed on days 1,2,3,4,7,14 postoperatively.The development of PCO was assessed by slit-lamp biomicroscopy at 12 weeks, and photographed by slit-lamp camera . Meanwhile, the histological observeation was performed using the light microscope and electronic microscope at 12 weeks postoperatively .The expression of ki67 was identified through immunohematological staining .The data were analyzed statistically by SPSS11.5 for window.Result1,The conjunctival and corneal reaction to the surgery.All eyes had conjunctival congestion during the first 5 days postoperatively , disappeared after 1 week. The cornea had mild to moderate edema ,which disappeared spontaneously after 3 to 4 days postoperatively. On the 1th day, the number of eyes that had edema had no significant difference.(Pï¹¥0.05)2,The anterior chamber reaction to the surgery.The anterior chamber had the different degrees of aqueous flare , which disappeared after 7 to 10 days postoperatively. On the 3th day,there was no significant difference between group A,B,C and D(Pï¹¥0.05) 3,The changes of intraocular pressure.The difference of the intraocular pressure was no significant at different interval time in each group. (Pï¹¥0.05)4,The number of eyes developing PCO at different time postoperatively.2 weeks postoperatively , PCO was began developing in group A. By 12 weeks, the number of the eye developed PCO in group A, group B, group C and group D were 8,5,3,1respectively(88.9%,55.5%,33.3%,11.1%). There was significant difference between the control group and the other groups at 12 weeks postoperatively(P<0.05).5,Histological examination.(1) At 12 weeks postoperatively, it was seen that multiplayer LEC proliferation in the center of the posterior capsule in the control eyes, while in the groupB,C and D, a few monlayers of LEC proliferation were seen.(2)The obvious pathologic changes were not found in the corneal endothelial cells, which appear regular hexagon in each group.(3)The expression of Ki-67: There was significant difference between the control group and the other groups at 12 weeks postoperatively(P<0.05). Conclusion1,Martine can significantly inhabit the proliferation of LECs and prevent the development of posterior capsular opacification by in rabbit eyes.2,The effect of Martine with the increase of the concentration(0.5g/l,1.0g/l,1.5g/l )was enhanced .3,As far as the concentration in this experiment was concerned, the toxicity to the anterior segment of the rabbit eye was not observed postoperatively.
