Radix Scutellariae is a traditional chinese medicine. Flavonoids baicalin, wogonoside, baicalein, wogonin are the main active components of Radix Scutellariae. Now the main extracting methods of Radix Scutellariae are decoction, reflux, etc. But those methods have low extractability. In order to make the best use of Radix Scutellariae effective methods should be seeked.In this paper, we study the extraction of active components(baicalin, wogono- side,baicalein, wogonin) from Radix Scutellariae by cellulase and pectinase. The concentration of baicalin, wogonoside, baicalein, wogonin were determined by HPLC. HPLC method was performed on methanol-acetonitrile-water-acetic acid (18:25:57:0.2) as the mobile phase at 30℃. The flow rate was 0.8 mL·min-1, and the detection wavelength was 275 nm. We also study the hydrolyze activity of four cellulases and three pectinases. The effects of buffer to material ratio, enzymatic hydrolysis temperature, the time of enzymatic hydrolysis, pH value of the enzymatic solution and the amount of enzymatic on the extraction yield of active components were investigated, the optium extraction technology of cellulase and pectinase was determined by orthogonal experiment. The optimum enzymatic hydrolytic conditions of celluase were determined as follows enzyme 25 U for 1 g material, pH 5.0, buffer to raw material ratio 20, temperature 50℃, and reaction time 7 h. The optimum enzymatic hydrolytic conditions of pectinase were determined as follows enzyme 15 U for 1 g material, pH 4.6, buffer to raw material ratio15, temperature 50℃, and reaction time 7 h. The optimum enzymatic hydrolytic conditions of celluase were determined as follows enzyme 15 U for 1 g material, pH 4.8, buffer to processing material ratio 30, temperature 50℃, and reaction time 7 h. The optimum enzymatic hydrolytic conditions of pectinase were determined as follows enzyme 5 U for 1 g material, pH 4.2, buffer to processing material ratio 20, temperature 50℃, and reaction time 6 h.we compare the effects of cellulase extraction, pectinase extraction,decoction, ultrasonic extraction, ethanol extraction of Radix Scutellariae.Compared with decoction, the cellulase extraction of raw material make yield of baicalin, wogonoside fall by 97.0% and 84.0%, baicalein, wogonin increase six and four times. The pectinase extraction of raw material make yield of baicalin, wogonoside fall by 96.8% and 80.3%, baicalein, wogonin increase five and three times. The yield of baicalin, wogonoside and baicalein, wogonin increase 16.3%,23.6% and 86.7%,90.9% from processing material by cellulase extraction. The yield of baicalin, wogonoside and baicalein, wogonin increase 11.7%,11.8% and 77.3%,81.8% from processing material by pectinase extraction.
