Donkey-hide glue and Cattle-hide glue are the tonification medicine with a history of 2500 years in our country.They have many pharmacological activities and are widely used for the blood increasing and hemostasis.The donkey-hide glue mainly uses in the anemia which is caused by many kinds of reasons;The cattle-hide glue is mainly used in blood loss.The quality control and evaluation of this two kinds of glue is mainly by character identification.Although Ch.P has a regulation of glycocine and the total nitrogen content,it can not react the real quality of donkey-hide glue.So it's an important task to establish a system for quality control and evaluation of glue as the donkey-hide glue and cattle-hide glue is more and more population and can make it enter the world's market as medical material today.Chromatographic fingerprints technique is a useful method in the identification of herbal samples or their products,and has been widely used for studying traditional Chinese medicines(TCM).It is an advanced technology for the quality and evaluation; CD spectrum is mainly used in the research for the three-dimensional of the organic compound,and it also can analyse the chiral compound in the complex matrix;The technology of HPLC-MS has a high ability of separation,selectivity and sensitivity. It also can provide the molecular mass and the structure information.A review on the development of research about chemical constituents and pharmacological activities of the donkey-hide glue was presented in this paper.It can give us a detailed understanding about glue as a medical material.Our research is for the quality control and evaluation of the donkey-hide glue and cattle-hide glue.It mainly include two parts.Firstly,we studied the chromatographic fingerprints of the water-soluble constituents in Dong E donkey-hide glue,Dong E Zhen donkey-hide glue and cattle-hide glue.We also compared the differences of tyrosine and phenylalanine in three kinds of glue.We used acetonitrile and water as mobile phase with a gradient elution.The wavelength is 205nm.HPLC separation was performed at 25℃.The fingerprints of the water-soluble constituents in three kinds of glue were gathered;Simultaneously, we quantitated the tyrosine and phenylalanine in different glues by external standard method.The result showed that there were 19 common peaks in Dong E donkey-hide glue,17 common peaks in Dong E Zhen donkey-hide glue and 18 common peaks in cattle-hide glue.The method validation met technical standard of fingerprints.The quantitated result showed that the content of tyrosine and phenylalanine in different glue are also different.Secondly,we identified the Dong E donkey-hide glue and its spurious breed by CD and HPLC-MS.The CD spectrum showed a big difference between the quality goods and spurious breed,the difference possibly was created by the structure and the content of the compound in the samples.We used the technology of HPLC-MS to screen the molecular mass of compound in Dong E donkey-hide glue and its spurious breed.The result showed the molecular mass and the structure information of the compounds in quality goods and the spurious breed.CD and HPLC-MS provided a new method for the identification of donkey-hide glue,and it also establish the foundation for the structure analysis of the compounds in glues.
