Background:Laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS) technique provides important technique platform for Chinese medicine proteomics with protein/peptide as main components.Objective:Analyzing water soluble protein/peptide from seven kinds of the traditional Chinese medicine(donkey-hide gelatin、deerhorn glue、tortoise-shell glue、earthworm、antelope horn、membrane of chicken’s gizzard、pangolin)primaryly by means of proteomics to institute the protein/peptide mass fingerprinting.Methods:By means of dialysis-desalination-freeze drying with saturated sulphuric acid amine sediment,we got the water soluble protein/peptide from the seven kinds of traditional Chinese medicine mentioned above, and analyzed their Mr-interval 1500-13000Da protein/peptide distribution and their relative molecular weight with Ciphergen Biosystems Inc. ProteinChip(?) Biology System (American PBSⅡ+) and the combined NP10 array, Surface enhanced laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS) technique.Results:There were 38 significant humps of water soluble protein/peptide relative molecular weight be obtained from donkey-hide gelatin. Comparing the protein/peptide and amino acid relative molecular weight, there were possibly 9 kinds of meaningful protein/peptide. There were 27 significant humps of water soluble protein/peptide relative molecular weight in deerhorn glue, and there were possibly 16 kinds of meaningful protein/peptide. There were 6 stable humps of protein/peptide relative molecular weight in tortoise-shell glue, and there may be 6 kinds of meaningful protein/peptide. There were 29 significant humps of water soluble protein/peptide relative molecular weight be obtained from earthworm. Among them, crude and dried earthworm had 17 humps of meaningful relative molecular weight, and processed earthworm had 12 humps. There was an amino acid residue among multiple adjacent protein humps, and multi-groups of relative molecular weight of crude and processed earthworm were extremely similar, indicating these peptides might be the same peptides. There were 25 humps of water soluble protein/peptide relative molecular weight in antelope horn. Among them, decoction group had 14 humps, and cold infusion group had 11 humps. Compareing the peptides,we found some disparity and the relative molecular weight of some amino acid residue were approximate.And the multi-group relative molecular weight of decoction group were extremely close to cold infusion group, so these pept ides might be the same peptides. There were 63 humps of water soluble protein/peptide relative molecular weight in all membrane of chicken’s gizzard, the dried product present 32 humps,while the processed product present 31 humps. There were 20 meaningful humps of water soluble protein/peptide relative molecular weight in pangolin.Conclusion:By means of protein array and laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS) technique, we got the peptide mass fingerprint (PMF) of the seven kinds of the traditional Chinese medicine,and these can serve as the digitizated quality control standard of them; and we got the reliable information for further isolation、purification and verification of the active protein/peptide of them.
