Immunohistologic Analysis Of Renal Apoe Expression In Lupus Nephritis

OBJECTIVE:To investigate the renal expression of ApoE in human lupus nephritis (LN)and its relationship with renal damage.METHODS:Renal expression of ApoE was examined in 40 LN patient biopsy samples by immunohistochemical staining.Using light microscopy to investigate hematoxylin Yihong(HE)staining kidney,and count the number of glomerular cells.The relationship between ApoE levels and renal pathologic changes,renal injuries were analyzed.RESULTS:Compared with normal controls,there was a marked up-regulation of ApoE expression in glomerular and tubular tissues.There was significant difference between them(P＜0.01).The expression of ApoE in glomerular was no stastistic difference between classⅡand classⅢ(Q value:2.7385,P＞O.05).The expression of ApoE in tubular was no stastistic difference between classⅡand classⅢ(Q value:2.5196,P＞0.05).There was significant difference in the expression of ApoE in glomerular in classⅡand classⅣ(Q value:21.2969,P＜0.01).There was significant difference in the expression of ApoE in tubular in classⅡand classⅣ(Q value:11.4845,P＜0.01).There was significant difference in the expression of ApoE in glomerular in classⅢand classⅣ(Q value:12.8918,P＜0.01).There was significant difference in the expression of ApoE in tubular in classⅢIand classⅣ(Q value:5.2874,P＜0.05).Spearman relationship analysis showed that the expression of ApoE in glomerular were positively related to the glomerular cell proliferation,renal histological score activity index and SLEDAI in LN(R value:0.9352,0.7990,0.7469,P＜0.01),and negatively related to serum creatinine clearance rate(R value:-0.6495,P＜0.01).The expression of ApoE in tubular were positively related to the renal histological score activity index and SLEDAI(R value: 0.8139,0.7848,P＜0.01),and negatively related to serum creatinine clearance rate(R value: -0.6714,P＜0.01).CONCLUSION:Only weak ApoE expression was found in normal renal tissue.The expression of ApoE was higher significantly in LN patients.The expression of ApoE was higher in classⅣthan classⅡand clssⅢLN.The expression of ApoE in glomerular and tubular was no stastistic difference between classⅡand classⅢ.Spearman relationship analysis showed that the expression of ApoE in glomerular and tubular were closely related to the renal histological score activity index and functional lesion in LN.The expression of ApoE may be associated with LN activity and progressive renal dysfunction,and can be used as an indicator of LN kidney disease in clinical.
