BACKGROUNDViral myocarditis (VMC) is an inflammatory disease of cardiac muscle which caused by viruses. The main histopathologic chages are degeneration and necrosis of myocardiocytes, infiltration of inflmamatoyr cells and fibrosis. The incidence of VMC has a tendency in recent years. Epidemiological and molecular biological studies indicate that Enterovirus especially the coxsackievirus b3 is the most frequently agent. almost nearly 50 percent viral myocardits are caused by coxsackievirus b3. So far as, the pathogenesis of VMC is not demonstrated completely and no specific therapy is found. Recent research reveal that viral replications, inflammatory media, injury induced immune systerm are related with the development and progress , and are closely to the prognosis of VMC. Cytokine are important immune active medium , which play an significant part in inflammation and sele-immune response induced by virus. And its expression correlated with the prognosis of VMC closely. Matrix metalloproteinases (MMPs) and their tissue inhibitor (infector (TIMPs) take part in decomposing collagen . The excessive expression and activity of MMPs or unfit MMPs/TIMPs proportion will induced myocardial collagen decomposed and replaced by the fibrous matrix absent of connective structure, which can result in myocardial collagen remodeling, expansive ventricle, thin wall, heart failure or myocardial . Through compared mortalities, histopathologic findings and expression of IL-6 and MMPs/TIMPs of various groups to explore whether Quduyangxinchongji reduced viral myocarditis, effects and mechanism of role, and provide evidence for treatment.Method1. To construct the model of acute myocarditis by intraperitoneal injection coxsackievirus B3 , then detecting and conforming by pathology. at the same time making an interfering by Quduyangxinchongji , to observe the surviral rate and myocardial histopathologic changes of every group , so as to investigate the effect of Quduyangxinchongji.2. Total RNA was extracted from myocardium for reverse transcription and proliferation of cDNA according to the manuscriptures' instruction. Data were analyzed by computer and the results were expressed in RQ of IL-6 gene toβ—actin.3. To observe the expression of MMP-2,MMP-9 and TIMP-1 gene by RT-PCR in viral group and treatment group ,so as to review the meachism of viral myocarditis.4. To observe the expression of IL-6,MMP-2,MMP-9 and TIMP-1 protein by Immunohistochemistry in viral group and treatment group , Data were analyzed by computer and the results were expressed in IOD of IL-6,MMP-2,MMP-9 and TIMP-1.Result1. the general condition of miceIn contral group, there are no obviously changes then in viral myocarditis group the mice display fewer action ,low dispirits .when treated with quduyangxinchongji , the surviral rate of mice are raised, the myocardial pathological are improved , the inflammatory cell inflatrate are decreased .It has manifest improve in myocardiaJ pathologic changes . 2. changes of myocardial pathological:It shows different pathological changes in viral myocarditis group. there are considerable inflammatory cell inflatrate and myocardial degeneration in viral myocarditis well no different changes in contral group . it displays lower inflammatory cell in viral myocarditis treated with Quduyangxinchongji.3. the viral titerThe viral extract in myocardium are raised in viral myocarditis group and then descent after treating with the drug, the viral titer in myocardium are remarkably lower , this comparing has statistical differences. The difference was significant in statistics (P<0.05).4. the detection of the expression of IL-6 mRNA and proteinThere are strong expression of IL-6 mRNA and protein in viral myocarditis group than in contral group. reduced. Levels of myocardium IL-6 mRNA and protein were significantly decreased in mice given Quduyangxinchongji compared with model group mice. The difference was significant in statistics (P<0.01).5. The influence on the expression of MMP-2,MMP-9 and TIMP-1 mRNAand protein that treated with quduyangxinchongjiThere are strong expression of MMP-2,MMP-9 1mRNA and protein in viral myocarditis group than in contral group. There are no different changes of the expression of TIMP-1 mRNA and protein . Levels of myocardium MMP-2,MMP-9 mRNA and protein were significantly decreased in mice given Quduyangxinchongji compared with model group mice. The difference was significant in statistics (P<0.05).Conclusion1. This study suggests that Quduyangxinchongji can protect the viral myocarditis of mice. Quduyangxinchongji enhanced the surviral rate , decreased the viral titer, lighten pathological changes . 2. The quduyangxinchongji decreased the expression of IL-6 mRAN and protein in viral myocarditis, inhabited the inflammation response moderately, and decreased the damage of myocardial.3. The quduyangxinchongji decreased the expression of MMP-2,MMP-9 mRNA and protein in viral myocarditis ,thus prevent expansive ventricle, heart failure.
