objective: To investigate the effects of Xin'an granules on the expression of Interleukin-6(IL-6)?Tumor necrosis factor –?(TNF-?)and CVB3 mRNA in viral myocarditis(VMC)rats.Methods: 1.60 healthy male BALB/c rats were randomly divided into a normal group,a model group,a ribavirin group(positive drug,0.02 g·kg-1·d-1)and the groups of high,medium and low-dosage Xin'an granules(12,6,3 g·kg-1·d-1),once a day with 10 rats in each group.For the normal group,0.15 ml of normal saline was intraperitoneally injected into rats,while for the other 5 groups,CVB3 Nancy liquid with virus(0.15 ml 10-8 TCID50·ml-1)was injected.The rats in the drug administration groups were given corresponding drugs ig,while those in the normal group and the model group were given normal saline ig,for 7consecutive days.2 Observation index :(1)The general status of mice,the number of deaths in mice,and the survival rate were observed;(2)After successful modeling,all the rats were killed by breaking the neck after weighing the weight and the hearts of the rats were obtained through thoracotomy with asepsis.Clean the blood on the surface of the heart with filter papers.Then the cardiac muscle were dyed with HE for pathological rating myocardial pathological score rating;(3)CVB3 mRNA express of thecardiac muscle was detected by realtime-PCR and the level of protein TNF-?and IL-6 were detected with Western-blot.Results: 1.General status: the groups with virus injection manifested sluggishness,poor appetite,irritability and diarrhea,etc of different levels,compared with the normal group.the symptoms in model control group occurred quickly and were severe,the above-mentioned symptoms were obviously relieved in the group of feeling particles and ribavirin group,among which the high dose group of Xinan granule had the least symptom.2.Result of HE dye: the myocardial cells are in alignment in normal group without inflammatory cells,necrosis or swelling bleed myocardial cells infiltrating around the blood vessel.In model control group,the myocardial cells were disarranged,with interstitial edema being found and a large number of inflammatory cells infiltrating.3.compared with normal group,the death rate,CI,and Heart pathological ratings increased obviously(P<0.05)in model group.Compared with model groups,the death rate,CI,and Heart pathological ratings decreased obviously(P<0.05)in Xin'an granule and ribavirin groups.4.compared with normal groups,the express of CVB3 mRNA in cardiac muscle tissue increased obviously(P<0.05)in model group and compared with model group,it decreased obviously in Xin'an granule and ribavirin groups,most significantly in Xin'an granule groups(P<0.05).5.compared with normal groups,the level of TNF-? albumen and IL-6 albumen in cardiac muscle tissue increased obviously(P<0.05)in model group and compared with model group,that in Xin'an granule and ribaviringroups decreased(P<0.05),most obviously in high-dosage Xin'an granule group(P<0.01).However,compared with ribavirin group,the difference in Xin'an granule is not significant(P<0.05).Conclusion :1.Xin'an granule can reduce cardiac damage caused by viral myocarditis.2.Xin'an granule has obvious inhibiting effect on CVB3 viral replication in rats with acute Viral myocarditis.3.The effect of Xin'an Granule on the myocardium of mice with viral myocarditis may be related to the inhibition of viral replication and down regulation of the expression levels of TNF-and IL-6.4.the expression level of CVB3 mRNA,TNF-? and IL-6 albumen tended to vary synchronously in cardiac muscle tissue of the rats with Viral myocarditis.5.Xin'an granule showed a significant effect in treating acute Viral myocarditis in rats and the effect is dosage-related.
