AIM: To research this coexistence condition between the classical nerves transmittance acetylcholine and TH,CGRP in the facial nucleus of rats. And then, to research their distribution characteristic and form characteristic under this coexistence condition in the facial subnucleus of rats. And, to research this coexistence condition between TH and CGRP in the facial nucleus of rats.And, we studied their distribution characteristic and form characteristic between TH and CGRP. At the same time, we discuss meaning of coexistence in the facial nucleus of rats. We establish theoretical fundamentals in order to search for approach of curing hemifacial spasm on basis of"nucleus theory".METHODS: Both immunohistochemistry and immunofluorescence are used to study the distribution of neurons and fibers which are positive to tyrosine hydroxylase. And, we explore and discuss meaning of coexistence between acetylcholine and the two neuromodulators in the facial nucleus of rats.RESULTS: (1) We found TH-IR positive neurons in the facial nucleus of rats. Their neurons bodies and fibers show brown. The cells showed a variety of forms.The neurons show mainly form with long protuberance and they have forms like oval,spindle, ect. They have longer protuberance. They have multi-level branches. The protuberance connecting them to each other become them structure net-like. TH-IR positive neurons mainly located in ventral intermediate subnucleus (7VI), ventromedial subnucleus(7VM), lateral subnucleus (7L).(2) We also found plenty of TH and ChAT doubled labelled positive neurons in 7VM and 7VI. And, TH-IR positive nervous endings surrounded these doubled labelled positive neurons.(3) CGRP-IR positive neurons mainly located in dorsolateral subnucleus(7DL),dorsomedial subnucleus(7DM),7VM. we also found plenty of CGRP and ChAT doubled labelled positive neurons in 7DM,7L,7VL. CGRP positive nervous endings surrounded these doubled labelled positive neurons.(4) Also plenty of TH and CGRP doubled labelled positive neurons were found in 7DL and 7L. TH and CGRP doubled labelled positive neurons were surrounded and connected each other by neuronic protuberance and fibers. TH and ChAT immunofluorescence doubled labelled positive neurons have a lot of obvious branches and bifurcation.CONCLUSION: We found their coexistence condition between the classical nerves transmittance acetylcholine and, TH,CGRP in the facial nucleus of rats and between TH and CGRP through our experiments. And then, we may conclude that acetylcholine,TH,CGRP have mutual effect and adjustment. They adjust neuronic activities in the facial nucleus of rats.
