Objective:To investigate effects of islet cells exposed to different lever glucoses,and it's possible mechnisms.Methods:Kunming mice's pancreatic islet cell were isolated and divided into six groups:group 1(glucose 5.6mmol/L,G1),group 2(7.8mmol/L,G2),group 3(11.1mmol/L,G3),group 4(16.7mmol/L,G4),group 5(22.2mmol/L,G5),group 6(27.6mmol/L,G6)according to cultured condition.After 72 hours,the insulin's secretion were evaluated by the radio-immunity technique;The expression of IRS-2 was detected by the immunocytochemical techniques;Apoptotic cells were identified by Hoechst33342 immunofluorescence techniques.Results:(1)The expression of IRS-2 of G3 group was the highest,more than any other group(P<0.05),while the G4,G5 and G6 groups' were significantly decreased than G1group(P<0.05);(2)The apoptosis cells rate were increased along with the glucose's lever,the G2,G3 groups show no distinguished alterations with the G1 (P>0.05),while the G6,G5 and G4 groups were greater than each other(P<0.05)o (3)The G2,G3 and G4 groups' insulin secretion levers were significantly higher thanG1 group(P<0.05),while that of the G5 and G6 groups had showed no significant alterations with that of G1 group(P>0.05). Conclusion:11.1mmol/L glucose could up-regulate the expression of IRS-2 of the pancreatic islet cells.More than 16.7mmol/L glucose may decrease the expression of IRS-2 so that induce insulin resistance and disfunction of pancretic beta cell all so increase apoptosis,at last leading to the pathogenesy of diabetes.
