Objective:In this study,immunohistochemistry was used to investigate expression situation of ERCC-1 and LRP proteins in NSCLC specimen, ananlyzed its relationship with clinical pathological factors and the survival time after patient suffered surgical resection.The purpose was to study their roles in expression situation of NSCLC and provide new theoretical foundation for clinicaling treatment,judging prognosis of NSCLC.Methods:Samples of 46 NSCLC and 16 normal lung tissues were detected by SupervisionTMtwo-step method of ERCC-1 and LRP proteins expression. SPSS(13.0)statistical software was applied for data analysis.A p-value less than 0.05 were condidered significant.The correlation between the expression of ERCC-1 and LRP and the major clinical pathological characteristics of NSCLC were analyzed by Chi-square test.Postoperative survival period analysis were computed by the Kaplan-Meier method and compared by the Log-tank test.The prognostic relevance of ERCC-1 and LRP and conventional prognostic factors were analyzed by COX regression model.Results:1.The positive expression rate of ERCC-1 and LRP in NSCLC specimens was 54.3%and 82.6%,respectively Positive rate of ERCC-1 and LRP expression in normal lung tissue was 6.25%and 6.25%,respectively.The different positive rates between NSCLC and normal lung tissue had statistical significance.2.Expression rates for ERCC-1 in pulmonary adenocarcinoma and squamous adenocarcinoma were 64.2%and 55.6%,respectively and were significantly higher than that in squamous carcinoma 47.8%.It was also more highly expressed in the patients under the age of 60 than in those over the age of 60.3.the Lrp expression was significantly correlated with degree of tumor differentiation(P<0.05).4.Positive expression rate of ERCC-1 in high center and low histological grades significance.Positive expression rate of LRP in high center and low histological grades was,respectively the difference had statistical signifiacance.5.The histological grades lymph node metastasis status,TNM stages,LRP expression were correlated with prognosis.Patients with lower histological grades lymph node metastasis,and later TNM stages associated LRP expression singly or ERCC-1 and LRP their combinated expression were significantly suggested unfavourable prognosis.6.Multivariate analysis of prognostic factors in NSCLC patients was carried out by COXS regression model,found that lymph node metastasis status,TNM stages,LRP expression were risk factors which affect survival time of patients(p<0.05).Conclusion:1.The positive rate of ERCC-1 and LRP expression in was higher in NSCLC tissue than that in normal lung tissue.High positive rate of ERCC-1 and LRP expression in NSCLC provide a theoretical basis for neoadjuvant chemotherapy.2.ERCC-1 and LRP expression was not correlated with TNM stages and lymph node metastasis staus of NSCLC patients.But positive expression rate of LRP in high center and low histological grades was,respectively the difference had statistical signifiacance.3.Median survival period of patients with LRP positive was significantly shorter than that with LRP negative.The result showed that LRP expression were associated with short survival and poor prognosis,Patient of ERCC-1 positive expression were associated with longer survival than patient of negative expression,But it had not statistical signifiacance.4.There was the risk of decressing survival time of patients with LRP and the ERCC1 the two union expression.5.The lymph node metastasis status TNM stages,ERCC-1 and LRP expression were correlated with prognosis and they were independent risk prognostic factors.
