| Objective:To study the expression of COX-2,VEGF,bFGF and MVD in HCC,and evaluate the relationship of COX-2 and angiogenesis in HCC.To analyze the relationship of COX-2 and clinicopathologic factors and the survival time of patients,so as to provide theoretical basis for prevention,treatment and prognosis of HCC.Methods:Detect COX-2,VEGF,bFGF and MVD marked by CD34 in liver cancer tissue(52),latero-liver cancer tissue(52)and normal hepatic tissue (10)by immunohistochemistry method(SP).To analyze the relationship between COX-2,VEGF,bFGF,MVD and the clinicopathologic index of HCC by x~2 test and Spearman interclass correlation,single factor survival analysis by Kaplan- Meier,survival curve by Log-Rank test.To analyze The expression of COX-2 and other factors correlating with the prognosis by Cox proportional hazard regression model,These results were analyzed by SPSS 13.0 statistical software package,a P-value less than 0.05 were considered significant.Results:1.The positive expression rate of COX-2,VEGF and bFGF in HCC was(69.2%,65.4%,63.5%)respectively.The positive expression rate of that in HCC was significantly higher than that in latero-liver cancer tissue (42.3%,36.5%,34.6%)and normal hepatic tissue(0%)(P<0.05).The MVD in HCC(43.23±16.87)was significantly higher than that in latero-liver cancer tissue and normal hepatic tissue(26.98±8.57;13.70±5.44)(P<0.05).2.There are significant positive correlation between the expression of COX-2 and VEGF,bFGF,MVD in HCC(r=0.762,0.703,0.825;P<0.01).There are significant positive correlation between the expression of VEGF and bFGF in HCC(r=0.802,P<0.01).3.There were significant differences between the positive expression rate of COX-2 in PTNM stageâ… -â…¡group andâ…¢-â…£group,pylic cancer embolus and no(p<0.05).There are no differences between the expression of COX-2 and sex,age of patients,HBsAg,AFP,size,numbers,peplos and grades of tumor.4.There are significant correlation between the prognosis of patients the expression of COX-2 in HCC and the diamete of tumors(>5cm),number of tumors,pylic cancer embolus,PTNM stage,the positive expression of COX-2 in HCC and latero-liver cancer tissue(p<0.05). However,there are no correlation between the prognosis of patients and sex,age of patients,HBsAg,AFP,size,peplos and grades of tumor(p>0.05).5.Cox proportional hazard regression model shows:The size of tumor,PTNM stage,the positive expression of COX-2 in HCC and latero-liver cancer tissue were an independent risk factor affecting the prognosis of patients with HCC(p<0.05), otherwise the number of tumors was not(p>0.05).Conclusion:The positive expression rate of COX-2 in HCC was high. There are significant positive correlation between the expression of COX-2 and VEGF,bFGF,MVD.This means there are closely relationship between COX-2 and angiogenesis in HCC during the development of liver cancer.There are correlation between the expression of COX-2 and pylic cancer embolus,PTNM stage.the survival time of patients whose expression of COX-2 are significant positive in HCC and latero-liver cancer tissue,was short.The prognostic of was bad.It was very important to detect the expression of COX-2 for judgment the degree of malignancy and prognostic of HCC. |
