Association Between The Polymorphism Of XPD Lys751Gln And Clinical Phenotype And Response To Chemotherapy Of Non-Hodgkin's Lymphoma

Objective This study was aimed to explore the relationship between the single nucleotide polymorphism of XPD Lys751Gln and the clinical phenotypes and response to chemotherapy in patients with non-Hodgkin's lymphomas(NHL).Method Ninety-seven NHL patients,as residents of guangxi zhuang autonomous region of china,were enrolled into this study.Blood sample was obtained at diagnosis,The single nucleotide polymorphism of XPD Lys751Gln were detected using PCR-restriction fragment length polymorphism assay.Based on unconditional logistic regression analysis (using the ordinal-logistic model study when the dependent variate is ordinal data),the association between different gene types and the clinical phenotypes,as well as the recent effect of chemotherapy and prognosis were analyzed by comparing odds ratio(OR)with 95%confidence interval(CI).Result(1)The XPD Lys751Gln(T＞G)polymorphism of the ninety-seven NHL patients,Frequencies of TT,TG genotypes were 81.4%(79 cases),18.6% (18 cases),GG genotype was not observed. (2)According to the TT genetype,there are no significant difference among the frequency of TG genotype and the clinical phenotypes including the cell sources(T/B origin),malignant degrees(low/moderate/high aggressive),with/without B symptom,international prognostic index(IPI) groups(low risk,low intermediate risk,high-intermediate risk and high risk group),(P＞0.05).(3)According to the TT genetype,the frequency of TG genotype in patients obtained an objective response(CR+CRu+PR)was similar to those failed to chemotherapy(SD+PD),no significant difference was abserved(P＞0.05),which was also observed between patients survived≥2 years and those survived≤2 years.(4)According to the TT genetype,As to the frequency of TG genotype in subgroup of patients suffered grade 0,Ⅰ,Ⅱ,Ⅲ,orⅣleukopenia,There no significant difference was observed between these subgroups(P＞0.05).Conclusion There was no association between the polymorphism of XPD Lys751Gln and the clinical phenotypes(cell source,malignant degree, with/without B symptom,IPI index)as well as response to chemotherapy (objective response,two-years survival,and leukopenia toxicity of chemotherapy).
