Leucocyte resides in male gential tract. Almost all male semen have leucocyte. Physiologically, leucocyte is from epididymis and has important function in immune defence mechanism. Leukocytospermia affects approximately 10%-20% of couples in male infertility. Semen leucocyte often increases in infertile male, and affects semen quality. The mechanism of leukocytospermia is not clear. The reports of leucocyte and semen parameters are yet disagree.In the current study, several comparative studies were carried out to clarify whether semen leucocyte influence on semen parameters or not, and to study relationship of between UU, CT and semen leukocyte. Semen specimens were obtained form 416 subjects of male infertility. The sperm morphology analysis using semiautomated sperm morphology analyzer (ASMA) was performed. To analyze Ureaplasma urealyticum, the culture method was used. To analyze Chlamydia trachomatis, enzyme method was used. They were measured by corresponding reagent kit. To analyze leukocyte number of semen, the bengidine staining method was used. Results:1. In semen specimen of 416 subjects of male infertility, the specimen of semen leukocyte >1×10~6 is 25.7% of all male infertility.2. Abnormal liquation time cases of abnormal semen leukocyte group were significantly higher than that of fertile group and normal semen leukocyte group (P<0.001); Semen volume of abnormal semen leukocyte group were significantly lower than that of fertile group and normal semen leukocyte group (P<0.05); Abnormal viscidity cases of abnormal semen leukocyte group were significantly higher than that of fertile group and normal semen leukocyte group (P<0.01); Abnormal pH cases of abnormal semen leukocyte group were significantly higher than that of fertile group and normal semen leukocyte group (P<0.01).3. Sperm density, sperm viability, sperm vitility of abnormal semen leukocyte group were all significantly lower than that of fertile group and normal semen leukocyte group (P<0.001, P<0.01). The percentage of morphologically normal sperm of abnormal semen leukocyte group were significantly lower than that of fertile group and normal semen leukocyte group (P<0.01, P<0.05). The percentage of head defect, neck/mid defect, tail defect sperm of abnormal semen leukocyte group were all significantly higher than that of fertile group and normal semen leukocyte group (P<0.01, P<0.05). In sperm motility parameters, VCL, VSL, VAP, ALH of abnormal semen leukocyte group were all significantly lower than that of fertile group and normal semen leukocyte group (P<0.01, P<0.05). BCF of abnormal semen leukocyte group was significantly higher than that of fertile group and normal semen leukocyte group (P<0.05).4. There is no difference in seminal plasma fructose, neutral - a glycosidase between of abnormal semen leukocyte group and of fertile group, normal semen leukocyte group(P>0.05). Seminal plasma acid phosphatase activity of abnormal semen leukocyte group was significantly lower than that of fertile group and normal semen leukocyte group (P<0.05).5. Sperm acrosin activity of abnormal semen leukocyte group was significantly lower than that of fertile group and normal semen leukocyte group (P<0.05). The percentage of apoptosis sperm of abnormal semen leukocyte group was significantly higher than that of fertile group and normal semen leukocyte group (P<0.05).6. Ureaplasma urealyticum infected cases of abnormal semen leukocyte group was significantly higher than that of fertile group and normal semen leukocyte group (P<0.05). There is no difference in Chlamydia trachomatis infected between of abnormal semen leukocyte group and of fertile group, normal semen leukocyte group(P>0.05).All results showed that abnormal semen leukocyte can influence semen quality. Semen leukocyte make semen parameters decreased, and sperm quality decreased. Abnormal semen leukocyte can influence seminal plasma acid phosphatase activity, and make sperm acrosin activity decreased. Abnormal semen leukocyte can make the percentage of apoptosis sperm increased. There is related abnormal semen leukocyte and infection of Ureaplasma urealyticum.
