Objective:To investigate therapeutic effect of monomer Rg1,Rb1 of Panax notoginseng saponins(PNS)on liver fibrosis of experimental rats.Methods:Healthy Sprague-Dawley(SD)rats were randomly divided into 10 groups: group 1(Rg1 120mg/kg/d),group 2(Rg1 60mg/kg/d),group 3(Rg1 30mg/kg/d),group 4(Rb1 120mg/kg/d),group 5(Rb1 60mg/kg/d),group 6(Rb1 30mg/kg/d),group 7(PNS 60mg/kg/d)),group 8(Colchicine 0.2mg/kg/d),group 9(Sodium Chloride)and group 10(normal control).All groups were given CCl4 by intradermic to establish the model of liver fibrosis except group 10.At the same time,the rats of group 9 were injected saline(1ml/d)by intradermic;the others were given drugs once a day(the dosage as mentioned above).In the first week of injection,one rat was sacrificed randomly in each group;In the second and the third week,two rats(one male and one female)were sacrificed randomly.The liver of rats was taken in order to observe the process of liver fibrosis.During the experiment period,the general condition,behavior and weight changes of rats were observed.According to the pathological changes,the degree of liver fibrosis and inflammation were divided into 5 grades with HE staining,Masson's staining and Gomori's staining.And the ultrastructure change of the liver were observed by electron microscope.Blood of rats was dropped to measure liver function and serological index of liver fibrosis.In addition,the expression of TIMP-1, Collagen(α1)type I at mRNA level in liver were measured by semiquantitative Reverse Transcription Chain Reaction(RT-PCR).Result:①During 5 weeks,3 rats of group 4 and 1 of group 7 and 1 of group 8 died.The others are all alive.The general condition of rats of the groups of Rb1 was worse than that of group 10,and it is no obvious difference with that of group 9.The general condition of rats of the groups of Rg1,group 7,group 8 was better than that of group 9 and the groups of Rbl.②The bodyweight of all groups was different after experiment,some of the changes had a significant statistical difference(P<0.05).③The stages of liver fibrosis among all groups had significant statistical difference(P<0.05 or P<0.01);The inflammation degree among all groups also had significant statistical difference(P<0.05 or P<0.01).④The index of liver function among all groups had significant statistical difference(P<0.05 or P<0.01).The serological index of liver fibrosis among all groups had significant statistical difference(P<0.05 or P<0.01).⑤The stereology data of liver cell mitochondria among all groups had significant statistical difference(P<0.05 or P<0.01).⑥The expression of TIMP-1, Collagen(α1)typeⅠat mRNA level in liver among all groups had significant statistical difference(P<0.05 or P<0.01).Conclusion:1.The rat model of liver fibrosis induced by CCl4 is a simple,easy and cheaper experimental model.2.Panaxsaponin Rb1,the monomer of panax notoginseng saponins,is not effective on hepatic fibrosis and is not the effective ingredients of Panax notoginseng saponins which can be used in hepatic fibrosis.In addition,the Rb1 have toxic effect on liver tissue.It is should be attentioned.The other monomer Panaxsaponin Rg1 have antifibrosis effect.The antifibrotic effect of Rg1 on animal model was more obvious than that of PNS in some aspects,with less toxic effect than Rb1.Therefore,Rg1 could be regarded as a reasonable traditional Chinese antifibrotic agent.
