Objective: To establish the model of multiple cerebral concussion(MCC) in rats. To observe the expression changes on the tryptophan hydroxylase(TPH) in brainstem and the tyrosine hydroxylase (TH) in the basal ganglia and the septal area, to explore the changes of the tryptophan hydroxylase expression and the tyrosine hydroxylase expression which are related to the cognitive behavioural deficits, so as to know the neurobiological mechanism of cognitive behavioral deficits after cerebral concussion in adult SD rats.Methods: A self-made metallic pendulum-striker concussive device was used to duplicate the rat MCC model. 48 Sprague-Dawley rats were randomly divided into six MCC groups: 1d,2d,4d,8d,16d and 24d group(n=8,in each group). One control was used(n=8). Brain tryptophan hydroxylase expression and tyrosine hydroxylase expression was detected by using the Western Blotting analysis system.Results: The western blotting analysis revealed that the TPH expression in brainstem rised from 1st day and arrived the peak in 2nd day after injuries in rats. To contrast with the control group, the values from 2nd day to 8th day have the statistical meanings. And then ,the protein's expression descended little by little and got the level of the control group in 24th day. About TH,the protein expression in the basal ganglia went up in the whole and the peak appeared in 4th day after injuries in SD rats. To compare with the control group, the expression from 2nd day to 16th day has the statistical meanings. While the protein expression in the septal area upgraded in the whole and the peak appeared in 4th day after injuries in SD rats. The value show that the expression has a little descendent in 8th day and 16th day and then rose again in 24th day. To contrast with the control group, the expression of the protein in 4th day and 24th day has the statistical meanings.Conclusions: 1. The brainstem TPH expression ugraded after the multiple cerebral concussion. 2. The basal ganglia and the septal area TH expression rose after the multiple cerebral concussion. 3. To combine with previous study in this item,the author supposes the cognitive deficit possiblely correlated with the brainstem TPH expression and the basal ganglia and the septal area TH expression.
