Objective:To establiish a model of the glioma of C6/SD mice and base on the model, purpose to explore the mechanism quantity effective and chronergy of the apy of artemether(Art)to glioma of C6 mice.Method:To build the model of glioma of C6/SD mice by the way of HE staining and flow cytometry to study the proliferathve kinetics and appotplis index under give Art to c6 neurospingioma in five body by different dose and phase.On the model1.Quantity effective:To dividen the SD mice(weight:250g-300g)to three groups: The experiment group(40)The control group(10)and The blank group(10).then according to the requestment of the ex-perimentation to divide the experiment group into 4 Sub-group(25mg/kg)For the low dose of Art,middle dose(50mg/kg)high dose(50mg/kg)and give chalybeate(1.5mg/kg)into middle dose.The contorlgroup is CCNU(10mg/m~2)and the biank groupis tales dose of NS.2.Chronergy:To divide SDmice into experimental group(50)and blank gyoup(10)and feed the C6/SD cultivated glioma by Art(50mg/kg)and chalybeatas by interral of 4h 6h 8h 12hand24h.Feed the blank group the tale dose of NS.Then to oberserve the proliferathve kinetics of C6 mice of neurogliocytoma and existence sitnation of each group.Results:1.Quantity effective:after plants tumor cell successfully to feed the Art of different dose observe and arrange each group from the best to worst,the oder is:the group of middle dose added chalybeatas has the equal effective of the group of CCNU then is the group of high dose,middle dese,low dese.the last is blank dose. HE staining the group of middle middle dose added chalybeatas and the group of CCNU have the greatest increasing in the area of C6 neurosponghoma necrosis.Then, the order from grenter to tiny:be the group of high dose.Idle dose,low dose,blank group.Detection of flow cytometry:It can be found the order that the percentage of apoptosis of C6 neurospongioma of each group arrange from high to low is according to the abved.2.Chronery:On the base of the first port choosen the middle dose of Art and added the chalybeatus and obeseve extince condition of the feeded mice the order from the best to the wrost is the geoup the 6h 8h 24h 4h 12h and blank group.HE staining the condition of area of cell necrosis of C6 neurosp ongioma is like aboveDetection of flow cytometry It can be found the order that the percentage of apoptosis of C6 neurospongioma of each group arrange from high to low according to the aboved.3.The detection of flow cytometry showed that there is no of signifficent of Art on the cell cycle of C6 neurospongioma.Conclusion1.We can observe the generation condition from the different angle though the HE staining and the diction of flow cytometry.2.There is depended relation slip of dose on the the effect of Art cure the brain glioma but on the condition of combination chaiybeatus the depended relationship can be to weaken at same time,it accelerate the induction of the apoptosis of the brain glioma to extend the extinction period of the glioma SD mice.3.We can reach the inhibitory action though feeding the Art and chalybeatus to mice by 6h intermission,and can extend the life spea of the brain glioma mice.4.The major mechiacsm of Art the to cause in hibitory action to C6 neurospongioma.the break of the neurospongioma DNA then occurrce the programmed appotisis. 5.There is no signifficent influence of Art tale effect on the cell cycle of neurospongioma.
