Research On The Inhibition Effect Of Artemether On The Glioma Cells In Vitro

Research on the inhibition effect of artemether on the glioma cells in vitro.Obejective:To study on whether the Art can induce the glioma cells C6 and U251 to be apoptosis.Metheods:1. Cells culture, for U251 cells:The U251 and C6 cells were supplemented in the culture medium(RPMI1640 for U251 cell/DMEMfl2 for C6 cells) containing 10% foetal calf serum, 100unit/ml penicillin and streptomy. Under the condition as below: 37℃, saturated humidity, and 5% CO2. Renew the medium every 3-4 days for U251 cells and evey 2-3 days for C6 cells. We divide the cells into different groups, the control group with no Artemether. All groups with Artemether ranged from 25-800umol/l(25,50,100,200,400,800 umol/1) with the treatment time for 24h,48h and 72h.2. Using the MTT assay to test the inhibition rate of artemether to the C6 and U251 cells, get the OD value, compute the IC50. 3. Study the cell cycle change by using the FCM. 4. Detect the apoptosis bodies by using the Hoechst33258 assay. Observe the apoptosis of C6 cells and U251 cells.Results:1. Artemether can inhibit the proliferation of C6 cells and U251 cells, The inhibition effect enhance with time and the dose of Artemether. By the MTT method, the IC50 values for C6 cells were 195.08±3.27umol/l,119.64±4.06umol/l,87.84±0.93umol/l respectively by the treatment time on 24h,48h and 72h. And 849.28±25.89umol/l,518.93±32.83umol/l,172.99±6.06umol/l for the U251 cells. 2. In the FCM system the apoptosis rate for the C6 cells were 7.95%,22.01%,31.22% in three time groups(24h,48h,72h) under the Artemether concentration in 200umol/l, and 4.55%,12.82%,24.22% for U251 cells under the Artemether concentration in 400umol/l.3. Under the Artemether concentration in 200umol/l for C6 cells and 400umol/l for U251 cells, we find the apoptosis bodies both in the two kind of cells by using the Hoechst33258 assay.Conclusions:1. Artemether can inhibit proliferation of both C6 cells and U251 cells, this inhibition showed a positive correlation with the processing time and the dose of Artemether.2. Artemether can make an influence in the cell cycle of C6 cells and U251 cells, blocking them in the GO/G1 phase and induce their apoptosis.
