It has been suggested that Aggressive periodontitis(AgP)is one of the main reasons that causes the healthful young people to lost their teeth,which has a genetic background and family aggregation.This disease is developed very rapidly.The classification of Aggressive periodontitis is ambiguity and the etiology and pathogenesis of Aggressive periodontitis are not clear until today.Not with standing, some microorganisms with special virulent factor could cause AgP.Currently,it is considered that the etiopathogenisis of AgP includes predisposing factor of host, environmental factor and pathogenic microorganism and the predisposing factor of host is decided by related genes.Evidences also indicate that Aggressive periodontitis may be involved with dysfunction of the cellular immune responses.Previous researchers have found that two groups genes(up-regulation and down-regulation)which could level express differently in AgP group compared to normal group by using microarray technology.Here we selected CYP2A7 from down-regulation group to detect the RNA transcriptional expression level in AgP,chronic periodontitis(CP)and normal control groups by using FQ-PCR and then analyzed it by bioinformatics.In this study,blood from peripheral vein were obtained from AgP patient,CP patient and healthy subject.Then the leucocytes were isolated from those blood. The total RNA was extracted from the leucocytes and cDNA was converted by using reverse transcription enzyme.The expression level of the CYP2A7 gene was examed by using quantitative PCR technique.Furtermore,bioinformatics analysis was employed to analyze this gene.From those experiments,we found that the RNA transcriptional expression levels (Ratio=0.43)in AgP patients were significantly lower than that in normal subjects. The difference levels are consistent with the results from microarray screening.In addition,we found that the RNA transcriptional expression levels(Ratio=0.99)in CP patients were similar to those in the health subjects.CYP2A7 is predicted by bioinformatic analysis as one of the members of CYP450 that has the most similarity to CYP2A6.So,we suggested that CYP2A7 may be an oxidation reduction enzyme and participate in the metabolism of exogenous compounds and lipid in leukocytic cells.The unusual expression of CYP2A7 could induce dysfunction of leukocytic cells.The down-regulation of CYP2A7 gene expresson in AgP patients also suggested that this gene may be associated with the genetic predisposition to aggressive periodontitis.Further experiments need to be carried out to confirm these assumption.The inside understanding of the gene expression of CYP2A7 would help us better learn the etiopathogenisis and pathogenetic of AgP.
