Objective:To investigate the expression of NRP-1 in colorectal adenoma and adenocarcinoma,its role in tumor formation and the mechanism of interaction between NRP-1 and VEGF.Methods:Steptative-peroxidase-biotin immunohistochemistry technique was used to detect the expression of NRP-1 and VEGF in sixty-five colorectal adenoma specimens and sixty-six colorectal adenocarcinoma specimens.SPSS13.0 statistical package was used to analyze the relationship between the expression of NRP-1,VEGF and the clinicopathological festures in colorectal adenoma and colorectal adenocarcinoma.Results:(1) Immunohistochemistry result of NRP-1 and VEGF in colorectal adenoma and colorectal adenocarcinoma was similar, that is: a stronger stain was observed in colorectal adenocarcinoma tissue (main intensity of staining ++-+++) compared to colorectal adenoma tissue (main intensity of staining +-++). All the differences of NRP-1 and VEGF between the two tissues were significant (NRP-1:P<0.05,VEGF:P<0.05).(2)The expression of NRP-1 and VEGF in colorectal adenocarcinoma was significantly correlated with differentiation and Dukes Staging(P<0.05);but without age,gender,and tumorlocation(P>0.05).(3) The expression of NRP-1 and VEGF in colorectal adenoma was significantly correlated with histological type and atypia(P<0.05).(4) There was a positive correlation between the positive expression of NRP-1 and VEGF in sixty-six colorectal adenocarcinoma specimens(r=0.576,P<0.05).Conclusions:(1)The expression of NRP-1 in colorectal adenocarcinoma tissues is much higher than that in colorectal adenoma tissues;It suggests that they may play an important role in colorectal adenocarcinoma formation and development.(2)The expression of NRP-1 in colorectal adenocarcinoma was significantly correlated with differentiation and Dukes Staging ,which suggests it may be an important factor related to poor prognosis and an powerful predictor in evaluating tumor malignant biological behaviour.(3)The expression of NRP-1 in colorectal adenoma was significantly correlated with histological type and atypia,which suggests it may participate malignant transformation of colorectal adenoma and may be a monitor factor related to colorectal adenocarcinoma formation.(4)There is a positive correlation between the positive expression of NRP-1 and VEGF in colorectal adenocarcinoma which suggests NRP-1 can reinforce the biological effect of VEGF and promote angiogenesis,tumorigenesis and development in colorectal adenocarcinoma.Detection of NRP-1and VEGF may be significance not only to know the mechanism of interaction between neuropilin-1 and VEGF, but also to provide some experimental and clinical evidences for anti-angiogenesis treatment. |
