Objective: In order to investigate the expression and correlation of M1 macrophage marker CD86 and M2 macrophage marker CD163 in human colorectal adenocarcinoma and colorectal adenoma.Through the construction of rat colorectal tumor model,to further verify the expression of CD86 and CD163 in colorectal cancer tissue,and to explore the effect of celecoxib intervention on the expression of both,in order to provide some theoretical basis for celecoxib prevention and treatment of colorectal tumor.Methods:1.Immunohistochemical method was used to detect the expression of CD86 and CD163 in 11 groups of human colorectal adenocarcinoma tissue(adenocarcinoma group),normal colorectal tissue(normal group)and colorectal adenoma tissue(adenoma group).Western blot method was used to detect the expression of CD86 and CD163 in 11 groups of colorectal adenocarcinoma tissue(adenocarcinoma group),matched paracancerous tissue(paracancerous group)and colorectal adenoma tissue(adenoma group).Furthermore,statistical software was used to analyze the correlation between them and the relationship between them and clinicopathological features.2.18 SD rats were randomly divided into control group(Control),model group(Model)and drug intervention group(Intervention).The rats in the control group were given intraperitoneal injection of normal saline(20mg/kg)every week,and the model group and drug intervention group were given intraperitoneal injection of DMH(20mg/kg)every week for 20 weeks.At the same time,the rats in the drug intervention group were given intragastric administration of 24mg/kg celecoxib every day for 36 weeks.The colorectal tissues of rats in each group were stained with hematoxylin-eosin to observe the construction of tumor,and the distribution and expression of CD86 and CD163 in colorectal tissues of control group and other two groups were detected by immunohistochemical method and Western blot method respectively.Results:1.The results of human tissue experiment:(1)the results of immunohistochemistry showed that the expression of CD86 in adenoma group and adenocarcinoma group was higher than that in normal group,while the expression of CD86 in adenoma group was significantly higher than that in adenoma group(P<0.05).The expression of CD163 in normal group and adenoma group was higher than that in adenocarcinoma group,but there was no significant difference between normal group and adenoma group(P>0.05).(2)The results of Western blot showed that the expression of CD86 in adenocarcinoma group and its adjacent paracancerous group was higher than that in adenoma group,while the expression of CD163 in adenoma group and paracancerous group was higher than that in adenocarcinoma group(P<0.05).(3)there was no significant correlation between the expression of CD86 and CD163 in colorectal adenocarcinoma(P>0.05),however,the expression level of CD86 was related to the CEA and lymph node metastasis(P<0.05),but not to sex,age,tumor size and tumor location(P>0.05).The expression level of CD163 was not related to sex,age,tumor size,tumor location,CEA level and lymph node metastasis(P>0.05).There was no significant correlation between the expression of CD86 and CD163 in colorectal adenoma(P>0.05),and there was no correlation between the expression of CD86 and CD163 and the sex,age,size and location of colorectal adenoma(P>0.05).2.The results of rat tissue experiment:(1)the results of rat model showed that there was no tumor formation in the colorectal tissue of 6 rats in the control group,and 38 tumor nodules were successfully induced in 6 rats in the model group.5 of the 6 rats in the drug intervention group successfully induced tumors and formed 12 tumor nodules.(2)the results of immunohistochemistry and Western blot showed that the expression of CD86 in the model group and drug intervention group was higher than that in the control group(P<0.05),but there was no significant difference between the two groups(P>0.05).The expression of CD163 in the control group and drug intervention group was higher than that in the model group(P<0.05),and there was no significant difference between the two groups(P > 0.05).Conclusion:1.In the evolution of "normal-adenoma-adenocarcinoma",the expression of CD86 is gradually up-regulated,while the expression of CD163 is relatively high in normal and adenoma tissues,and down-regulated in adenocarcinoma tissues。It is suggested that M1 and M2 macrophages may be involved in the development of colorectal adenocarcinoma..2.In colorectal adenocarcinoma,the expression level of CD86 is related to the level of CEA and lymph node metastasis.CD86 may be used as a biological index to evaluate the condition and prognosis of patients with colorectal adenocarcinoma.3.Celecoxib may play a role in tumor prevention by restoring the infiltration of M2 macrophages in colorectal tumors.
