Mucoepidermoid Carcinoma (MEC) of salivary gland is the most common primary salivary gland epithelial malignancy. Deregulation of cell cycle plays a key role in cell proliferation and tumor development and progression. Cyclin D1 is a critical factor in regulating G1-S transition, while Ki67 is often used to evaluate cell proliferative capacity due to its continuous expression in proliferating cells. Up to now, we know little about the relationship between cell cycle deregulation and MEC development.Objectives: The purpose of the present study is to investigate the expression of Cyclin D1 and Ki67 in MEC and assess their roles in the carcinogenesis and prognosis of MEC. Furthermore, we want to evaluate the possibility of using them as the biological markers of MEC for clinical arrangement and prognosis.Methods: The immunohistochemical method was used to detect the expression of Cyclin D1 and Ki67 in 34 cases of MEC and 10 cases of normal salivary glands (NSG). Data were analyzed using SPSS version 11.5 for windows.Results: Cyclin D1 expression could hardly be observed in normal salivary glands, but 73.53% of MEC showed positive staining. There was a significant difference between them (p<0.01). The expression rate of Ki67 in normal salivary gland and MEC were 10% and 85.29% respectively. There was a significant difference between them (p<0.01). The expression rate of Cyclin D1 in the high grade and the low grade of MEC were 57.89% and 93.33% respectively. There was a significant difference between them (p<0.01). The expression rate of Ki67 in the high grade group and the low grade group of MEC were 78.95% and 93.33% respectively. There was a significant difference between them (p < 0.01). Cyclin D1 positive expression could be recognized in all of recurrent cases, while only 68.79% of non-recurrent cases showed Cyclin D1 expression. There was a significant difference between them (p< 0.01) . Ki67 showed positive expression in all of recurrent cases and 82.76% of non-recurrent cases. There was a significant difference between them (p < 0.01). All of metastatic cases showed Cyclin D1 positive expression, but only 68.79% of non-metastasis cases showed Cyclin D1 expression. There was a significant difference between them (p<0.01). The expression of Ki67 in metastasis group and no metastasis group were 100% and 68.79% respectively. There was a significant difference between them (p<0.01). The expression of Cyclin D1 had a positive correlation with that of Ki67 (p<0.01).Conclusion: The expression of Cyclin D1 might play a certain role in the development and progression of MEC. Cyclin D1 and Ki67 expression also related with the recurrence and metastasis of MEC. Combined analysis of the expression of Cyclin D1 and Ki67 might be useful to make the diagnosis and prognosis of MEC.
