Objective: Through establishing the model of myocardial ischemia-reperfusion, to explore the protective effects and possible mechanism of erythropoietin in the process of myocardial ischemia -reperfusion injury in rats,and to provide theroial base for clinic application .Methods: 60 Wistar rats were randomly divided three groups: sham-operation(n=20), ischenmia–reperfusion(n=20) , EPO(n=20).Using multiplying channel electrocardiogram monitor ontinuously was to note the arrhythmia caused by reperfusion. Tracheal intubation was contactedUsingsmall animal breathing machine to assist respiration with positive pressure. Rats of EPO group were injected with EPO(5000U/Kg) in abdominal cavity, the others were injected with nomal sodium. The left anterior descending coronary artery was ligated for 30minutes and then loosed for 150minutes to establish the rat model of myocardial ischemia -reperfusion injury. During the processes,leadⅡ/ECG was traced continuously to note the arrhythmia caused by reperfusion;the levels of cardiac troponin(cTnⅠ) were detected ;the levels of Malondialdehyde (MDA) were detected.The myocardial apoptosis index of rats were detected. nd the changes of myocardial ultrastructures were observed.Results:Erythropoietin significantly reduced the incident of ventricle arrhythmia caused ischemia–reperfusion.Conclusion:Erythropoietin has protective efects on myocardial ischemia-reperfusion injury in rats,and the mechanism may be related to relieving the injury caused by oxygen free radical and calcium overload.
