| Objective:For providing theory basis,we fed rats with high fatty diets for causing nonalcoholic fatty liver disease(NAFLD),then observed the function of total Saponins of Panax notoginseng (tPNS) against NAFLD to probe into the influence of different dosages on insulin resistance (IR),serum leptin(LP) and leptin receptor(leptin-R),and further more to study the relationship between IR and NAFLD,LP and NAFLD,leptin-R and NAFLD,to assess the treatment effect and to explain the path of tPNS treating NAFLD.Method:40 healthy male Wistar rats were randomly divided into two groups:normal group with 10 rats and model group with 30,they all were fed liberally.The normal group was fed with ordinary diets and the other was fed with high fatty diets.Three months later,selecting randomly 2 rats from normal group and 6 ones from model group to get their liver tissue separately and then to dye them with HE tinction to identify if the model was available.If it was successful, high fatty diets were stopped to feed the model group and were changed to ordinary diets.Then the remaining rats were divided randomly into 4 groups with 8 in every group.The situation liked this:the normal group was fed with pure water by 4ml·d-1 for 30 days,the model group was fed with pure water by 4ml·d-1 for 30 days,low tPNS dosage group with tPNS by 100mg·kg.(-1)·d-1 for 30 days,high tPNS dosage group with tPNS by 200mg·kg-1·d-1 for 30 days.One month later, we got rats' weight and wet liver weight to count the liver index,got rats' serum to inspect the consistency of LP and FINS by radioimmunoassay,to inspect the blood sugar and HOMA-IR under empty stomach,and took the liver tissue of rats to express the leptin-R by immunohistochemical and HE tinction.Result:1.High fat diets induced NAFLD model successfully.2.Compared to the normal group,the model group was higher in weight,liver index,every physiological and chemical index and leptin-R expressed by liver tissue,they were statistically different(P<0.05).The leptin-R of liver tissue in normal group expressed a few,it was weak positive in liver cell serum,compared to the normal group,the leptin-R of liver tissue in model group expressed a lot,it was brown strong positive in HSC and in sinus cell serum and cell membranes,they were statistically different(P<0.05).3.Compared to the normal group,the weight and liver index were great lower either in high tPNS dosage group or in low group,and they were statistically different(P<0.05).Compared to the normal group,the IR index and LP were great lower,they were statistically different(P <0.05) either.The high and low dosage tPNS together could cut down the amount of the leptin-R or of the normal group,and the high dosage group was more different(P<0.05).4.It was related positively among liver index,serum LP,insulin resistance index and leptin-R, and was statistically different(P<0.05).Conclusion:The tPNS is effective to treat the NAFLD,and the more dosage increases,the better effect gets.Dropping the liver index and serum LP,improving the insulin resistance and decreasing the leptin-R may be the paths to treat NAFLD. |
