The Changes Of The Platelet Membrane Glycoprote In CD62P And VWF In Rats After Acute Ischemia-reperfusion Of Hind Limb And The Interventional Effects Of Flurbiprofen

Objective To investigate the changes of the platelet activation and endothelial cell disturbance in rats after acute ischemia and reperfusion of hind limb.And to investigate the effects of Flurbiprofen on the expression of the platelet membrane glycoprote in CD62P and vWF.Method Part 1 of the experiment,eighteen rats were randomly divided into 3 groups:the sham operation group(N group);the ischemia 2h group(I₂);the hind limb ischemia 2h and reperfusion 3h group(I₂R₃ group).An animal model of acute hind limb ischemia and reperfusion was developed by clamping femoral artery of rats.Get the blood from the heart.Changes in the expression of CD62P of platelet membrane glycoprote and vWF in rats were observed following ischemia and reperfusion.Based on the Part 1 of the experiment,the hind limb ischemia 2h and reperfusion 3h plus the Flurbiprofen group(F+I₂R₃ group ) was added in the Part 2.Eighteen rats were randomly divided into 3 groups according to the different dosages of Flurbiprofen:the hind limb ischemia 2h and reperfusion 3h plus the Flurbiprofen 3mg group(F₃+I₂R₃ group);the hind limb ischemia 2h and reperfusion 3h plus the Flurbiprofen 6mg group(F₆+I₂R₃ group);and the hind limb ischemia 2h and reperfusion 3h plus the Flurbiprofen 12mg group(F₁₂+I₂R₃ group). Flurbiprofen was given from vena caudalis before clamped femoral artery.Changes in the expression of CD62P of platelet membrane glycoprote and vWF in rats were observed following Flurbiprofen treatment.Result The expression of CD62P of platelet membrane glycoprote and vWF in the hind limb ischemia 2h and reperfusion 3h group(I₂R₃ group) was significantly higher than that of the sham operation group(N group)(P＜0.05),while the expression of vWF in the ischemia 2h group(I₂) was significantly higher than that of the sham operation group(P＜0.05).There were no significant changes in the expression of CD62P of platelet membrane glycoprote among the I₂R₃ group and the sham operation group.The expression of CD62P of platelet membrane glycoprote and vWF in F+I₂R₃ group was significantly lower than that of the I₂R₃ group(P＜0.05).Conclusion Acute hind limb ischemia and reperfusion can increase the platelet activation and endothelial cell disturbance in rats,and increase the expression of CD62P of platelet membrane glycoprote and vWF in rats.Early Flurbiprofen treatment can inhibit effectively the expression of CD62P of the platelet membrane glycoprote and vWF,which may help to relieve reperfusion injury.
