Pocynum hendersonii is a perennial herbaceous plant from Poacynum Baill in Apoacynacease. It had been reported that the extract from the leaves of Pocynum hendersonii exerts various pharmacological effects including antihypertensive and reducing serous lipoprotein and antihematoblast collective induced by ADP as well as antioxidant effect.The Preliminary Phytochemical composition of Poacynum hendersonii leaves was tested. The results showed that the Poacynum hendersonii leaves mainly contains phenolic substances, tannic materials, flavonoids, anthraquinones, caridiac glycosides, lactones, coumarins as well as volatile oils. The extraction technology of total flavonoids in Poacynum hendersonii leaves by ultrasonic wave was studied through single factor experiments and orthogonal experiments. The results showed the optimal conditions for the extraction with ultrasonic frequencyâ… (26.32Hz),the treatment time 20 min, ethanol concentration 70% and the ratio of material to solvent 1:50.The primary extracts solution was obtained by the optimal conditions for the extraction and the percolation method. The primary extracts were separated into two fractions by systemic solvent extraction method with ethyl acetate and normal butanol. The two fractions were isolated by silica gel column chromatogram . finally the fractions were purified further by repeating Sephadex LH-20 column chromatography and repeating crystallization.The purity of compounds were examined primarily by TLC, at last the structures of compounds were identified by instrumental analysis means, such as IR, MS, 1H-NMR, 13C-NMR.One compound-Isoquercetin had been yielded in the experiment.
