Studies On Chemical Constituents Of Large-leaved White Cannabina

Pocynum hendersonii(Hook.f.) Woodson is a perennial herbaceous plant from Poacynum Baill in Apoacynacease , and mostly grows in Qinghai ,Xinjiang ,Xizhang and other provinces of China. It has been reported that the alcohol extract from the leaves of Pocynum hendersonii(Hook.f.) Woodson exerts various pharmacological effects including antihematoblast collective and antisenescent and antihypertensive and reducing serous lipoprotein as well as improving cordis function effects. The chemical constituents from leaves of Pocynum hendersonii(Hook.f.) Woodson were extracted, isolated, purified and finally identified in the experiment .The leaves of Pocynum hendersonii(Hook.f.) Woodson,a kind of crude materials used in the experiment , were supplied by Yabao pharmacy factory in Shanxi province , and were collected from Geermu region in Xinjiang province,the identification t of the experimental materials was performed by professor Jianping Gao, the director of crude drug laboratory from the Pharmaceutic School, Shanxi Medical University.The primary extracts was obtained by dipping the medicinal herb in alcohol solution which concentration was 75% at the room temperature.And then solvent was recycled after vaporized from the solution in the condition of low pressure.The primary extracts were separated into five fractions by systemic solvent extraction method, namely through petroleum ether,chloroform,ether acetate and normal butanol respectively, and then the petroleum ether fraction was separated by silica gel column chromatogram and was eluted by different concentration of petroleum ether-ether acetate; meanwhile the ether acetate fraction was separated by polyamide column chromatography and was eluted by different concentration of alcohol-water and ether acetate-methaol.Petroleum ether fraction was purified further by repeating silica gel column chromatography ,which was eluted by different concentration of petroleum ether-ether acetate and petroleum ether-acetone, and was finally purified by a means of repeating crystallization. The ether acetate fraction was purified further by repeating Sephadex LH-20 column chromatography , which was eluted by methaol and chloroform - methaol , and finally by a means of repeating crystallization.The purity of compounds were examined primarily by TLC and melting point along with and other physical and chemical property. The type of characteristic structure and framework can be judged by various colour reactions and IR , at last the structure of compounds were characterized by instrumental analysis means, such as 1HNMR and 13CNMR.
