Cedrus Trew of Pianaceae is generally called as Cedrus deodara, its pine needles is one of the 13 medicinal pine needles,which has a variety of pharmacological activities such as antiviral,anticancer,analgesic and so on.Firstly, in order to develop and use the pine needles of Cedrus deodara sufficiently, the thesis performed separation and purification of the chemical constituents in n-butanol extract of pine needles of Cedrus deodara by macroporous resin and silica gel and Sephadex LH-20 and polyamide chromatography column, and the structures were elucidated on the basis of physicochemical properties and spectral data( 1H-NMR,13 C-NMR and DEPT). The result is seven compounds were isolated, including 1-( 4’ – hydroxyl- 3’ – methoxyphenyl) – 2- [ 4 "-( 3 – hydroxypropyl)- 2" – methoxyphenoxy ]- 1, 3 – propanediol(1),(7S, 8R)-9, 9’ – dihydroxy- 3, 3’ – dimethoxy- 7, 8 – dihydro – benzofuran- 1’ – propanol ase neolignan – 4 – O – β – D – glucoside(2),( 7R, 8R)- 3’, 9, 9’ – trihydroxy – 3 – methoxy- 7, 8 – dihydro – benzofuran- 1’- propanol base neolignans – 9 – O – α – L – rhamnoside(3),( 6R, 9R) – 6 – hydroxyl – 3 – oxo – α – ionol – 9 – O – β – D – glucopyranoside(4),( 6R, 9R) – 3 – oxo – α – ionol – 9 – O – β – D – glucopyranoside(5), shikimic acid butyl ester(6), quinic acid butyl ester(7), which were all isolated from pine needles of C. deodara for the first time.Lastly, the thesis take Ginsenoside Rb1 as standard,the total saponins content in pine needles of Cedrus deodara was determined by UV spectrophotometric at 540 nm.UV detective wavelength is 540 nm.Select ethanol as extraction solvent,and with the index for the content of total Saponins of Cedrus deodara pine,and extraction time, solvent-solid ratio and concentration of alcohol were selected as effected factors based on single-factor experiment,which was designed by Box-Behnken central composite. Extraction technology parameters of total saponins of Cedrus deodara pine was optimized by response surface methodology.The results show that the content of total saponins had a good linear relationship in the range of 20 μg~140 μg, and the correlation coefficient was 0.9992, and the average recovery was 96.965%,which RSD was 1.96%. The optimizing ultrasonic extraction conditions were as follows: ethanol concentration was 73%,solvent-solid ratio was 13:1,and extracting time was 65 min,ultrasonic extraction time was twice.Under the optimum conditions,the average yield of total saponins in 3 validation experiments was 6.693 mg / g, which is close to the predicted value 6.50799 mg / g, and the correlation coefficient is 0.9942.The thesis enriched natural chemical compositions by studying the chemical constituents in n-butanol extract of pine needles of Cedrus deodara and laid on foundation for further comprehensive development and utilization of pine needles of Cedrus deodara through researching on extraction technology of total saponins.
