Objective To investigate the relationship between the expression of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2) mRNA and clinicopathologic behavior in human gastric cancer.Methods A total of 100 specimens resected was investigated by S-P immunohistochemical staining with an anti-MMP-2 monoclonal antibody and an anti-TIMP-2 polyclonal antibody and detected by reverse transcriptase polymerase chain reaction(RT-PCR).The expression of MMP-2 and TIMP-2 in various regions of the gastric caners tissues,paracancerous tissues and incisal margin normal tissues were detected and the relationship with clioicopathologic factors were analyzed. Results The difference of MMP-2 and TIMP-2 expression in gastric cancer tissues versus incisal margin normal tissues,gastric cancer tissues versus paracancerous tissues,and paracancerous tissues versus incisal margin normal tissues was statistically significant.The expression of MMP-2 and TIMP-2 protein in gastric cancer tissues was not related with sex and age.But the expression of MMP-2 was positively related with the differentiation,depth of invasion,lymph node metastasis,distant metastasis,and TNM stage while the expression of TIMP-2 was positively related with the depth of invasion,lymph node metastasis,distant metastasis,and TNM stage.And there was a positive correlation between the expression of MMP-2 and TIMP-2 in gastric cancer tissues.Conclusion MMP-2 and TIMP-2 may play an important role in the carcinogenesis,invasion and metastasis of gastric careinoma,expression of MMP-2 and TIMP-2 might be served as an objective indicator for estimating tumor progress and predicting occurrence of cancer cell metastasis.
