OBJECTIVE To explore the effects ofα-anordrin on androgen-dependent human prostate cancer LNCaP cells in vivo and in vitro.METHODS 1. SRB protein-staining method was used to investigate the suppression effects ofα-anordrin on androgen-dependent LNCaP cells. 2. Flow cytometry was used to detect the distribution of LNCaP cell cycle afterα-anordrin treated. 3. Male SD rats were used to investigate the effect ofα-anordrin on the growths of sex glands and accessory sex glands, and the testosterone(T) in serum were examined by the ELISA method. 4. The model of xenograft tumor in nude mice, which was established by inoculating androgen-dependent prostatic cells LNCaP into the back of nude mice subcutaneously, was used to investigate the suppression effect ofα-anordrin to the proliferation of tumor, and the T and prostate-specific antigen(PSA) in serum were studied by the ELISA method. 5. H & E stain was done to examine the difference in histopathology between control team and administration team. Immunohistochemistry of Ki67 and PCNA was done to study the effectα-anordrin on the proliferation of LNCaP cells.RESULTS 1. The proliferation of LNCaP cells were inhibited evidently after treated withα-anordrin, and the inhibition exhibited a dose-response and time-response relationship. 2. After treated withα-anordrin, the transition of LNCaP cell cycle was interrupted from G1 phase to S phase, the cells were arrested in G0/G1 phase. 3.α-anordrin could decrease the T level in serum and reduce the weight of male SD rat sex glands and accessory sex glands. After a period of time withoutα-anordrin treatment, the level of T in serum and the weight of sex glands and accessory sex glands could reverse to normal level. 4.α-anordrin could reduce the weight and volume of LNCaP xenograft tumor in male nude mice. 5.α-anordrin can decrease the T level in serum of nude mice and restrain the growth of sex glands and accessory sex glands in bearing LNCaP tumor nude mice.CONCLUSIONS 1.α-anordrin can significantly inhibit proliferation of LNCaP in vitro. 2.α-anordrin could influence the level of T in serum and the growth of sex glands and accessory sex glands reversibly in vivo by inhibiting hypothalamus - pituitary - gonadal axis. 3.α-anordrin can inhibit the growth of LNCaP xenograft tumor in nude mice evidently.
