Objective : To investigate the inhibition effects ofα-anordrin on human prostate cancer and sex glands growth and testosterone secretion of male SD ratMethods: 1.SRB staining method was used to detect the effects ofα-anordrin on androgen-dependent and androgen-independent human prostate cancer cell lines in vitro.2. A culture medium of no-phenol IMDM containing charcoal-sripped fetal serum was used to examine the effects ofα-anordrin on androgen-dependent prostate cancer cell in the presence or absence of R1881. 3. A CWR22RV1 prostate cancer athymic nude mice model was used to investigate the anti-prostate cancer effects in vivo.4. Normal and castrated rats were used to investigate the effects ofα-anordrin on sex and accessory sex organs weights gain, and total testosterone was examined.Results: 1.α-anordrin inhibited the proliferation of both androgen-dependent and androgen-independent prostates cancer cells in vitro, but its more effective in inhibition of androgen-dependent cells.2.α-anordrin could reduce the growth promotion effects of R1881 in L1A and CWR22RV1 cells.3.α-anordrin coud significantly suppress the growth of human prostate cancer CWR22RV1 in nude mice model.4.α- anordrin could inhibite the weight gain of normal rat testis, prostate, seminal vesicle and levator ani muscle, and reduced testosterone to castrate level.Conclusions: 1.α-anordrin was more effective in inhibition of androgen-dependent prostate cancer cells than androgen-independent cells. 2. In vitroα-anordrin has the ability to resist androgen's effect of stimulating development. 3.α-anordrin could inhibited the prostate cancer growth in vivo. 4.α-anordrin could inhibited the pituitary-sex gland axis.
