Objective To investigate the effects of NDGA, a general lipoxygenase inhibitor,on the proliferation and apoptosis of prostate cancer cell PC-3 and its relevant mechanism,which may provide a novel approach for the therapy of prostate cancer.Methods Cultures of prostate cancer cell were exposured to NDGA at the different concentrations of 0μmol·L-1,20μmol·L-1,40μmol·L-1,80μmol·L-1 and 160μmol·L-1 respectively.The effect of NDGA on growth inhibition and apoptosis inducement were determinated with morphometry,MTT assay and TUNEL.The activity of Caspase-3 were determinated with flow cytometry.Rusults By MTT assay and TUNEL assay,the hyperplasy of PC-3 were significantly inhibited in the 40μmol·L-1,80μmol·L-1 and 160μmol·L-1 groups(P<0.05), but the effcects were nonsignificant difference in 0μmol·L-1 and 20μmol·L-1 groups(P>0.05).The activity of Caspase - 3 in 40μmol·L-1,80μmol·L-1 and 160μmol·L-1 groups were significantly higher than other groups(P<0.05),but the expression of active Caspase-3 was nonsignificant difference in 0μmol·L-1 and 20μmol·L-1 groups.Conclusions The growth inhibition and apoptosis inducement could be taken by NDGA in prostate cancer cell PC-3 in a dose and time-dependent manner.The mechanism was that the high activity of Caspase-3 was caused by NDGA, which led to the apoptosis of PC-3.
