【Objective】To investigate the expression of the insulin-like growth factor- 1 receptor on the epithelium of mouse oxidative injury cataract and transparent lens, for giving the experiment basis to the generation of cataract.【Methods】1.Adopting peroxide oxidative injury method to build experimental cataract lens model: the adult mouse transparent lens were randomly divided into 2 groups: control group and experiment group. The control group content: Medium199 cultured liquid+penicillium 2×105 U·L– 1+ streptomycin 2.5×105 U·L– 1; The Experiment group content: the control group cultured liquid+ H2O2 (final concentration 200μmol·L– 1). There are 24h, 48h, 72h time-limited subgroups in every group, measure every subgroup lens transparence. Then measure expression of the level difference among adult mouse transparent lens and different time-limited cultured cataract lens epithelium from immunohistochemistry, when IGF-1R in all of four.2.Using immunohistochemistry to measure expressions of IGF-1R of transparent lens epithelium in different age groups.Newly born mouse, adult mouse and aged mouse are included.【Results】1.Successfully construct experimental cataract lens model by using lens culture technology of peroxide oxidative injury method. Mouse lens in the control group kept its transparence in lens culture technology and not apparently obvious change in 24h, 48h, 72h time-limited subgroups. As the time pass the turbidity spread out inexperiment group, turbidity appeared in 24-hour, 48 hours later the turbidity spread to the center, when in 72 hours lens are totally turbid. The difference between control group and experiment group are highly prominent in every time point (t-test, p<0.001). There was a statistically significance among the experiment subgroups(analysis of variance F=43.609 ,P<0.001)and between every two subgroups(LSD-Ttest,P<0.001),but not among the control subgroups(analysis of variance F=2.316,P>0.05)。From the experiment data we know as the time of peroxide injury were prolonged, the turbidity will be heavier.2.IGF-1R are concentrated to lens epithelium plasm. The expression was strong masculine in adult mouse transparent lens, 24h cataract group, median masculine, 48h cataract group, faint masculine, 72h cataract group, little even not expression. The difference among groups has a statistically significance(Kruskal-Wallis检验, P<0.001),and the correlation between the expression of IGF-1R and peroxide injury time, rs=-0.725,(P<0.001),i.e. As the peroxide injury time got longer,cataract got heavier, and the expression of IGF-1R had an decreasing tendency.In different age groups, the expression of IGF-1R of newly born mouse was strong masculine, adult mouse, median-strong masculine aged mouse, little even not expression. The difference among groups has a statistically significance(Kruskal-Wallis检验, P<0.001),and the correlation between the expression of IGF-1R and mouse age, rs=-0.859,(P<0.001),i.e. As the mouse age grows, the expression of IGF-1R had an decreasing tendency.【Conclusion】Expression decrease of IGF-1R may have something to do with the aging of lens and the development of cataract.
