Objective:To detect expression of the genes associated with TGF-β/Smad signaling pathways in RA patient and evaluate their effect in RA,evaluate their expressions in progress and outcome of RA patient,to investigate the ralation of those detected factors and the syndrome types of Traditional Chinese Medicine(TCM),to provide an important theory of diagnosis and treatment in RA.Methods:Apply nested and conventional SYBR GreenⅠFluorescent Dye real-time-PCR to detect expression of TGF-βRⅡ,Smad4 and Smad7mRNA.RNA was isolated from PBMC and reversed transcription into cDNA as temproteinaceous corneal degeneration.Design outer and inner primers by TGF-βRⅡ,Smad4 and Smad7 gene sequence.Detect TGF-βRⅡ,Smad4 and Smad7mRNA gene expression level in Real-Time-PCR using SYBR GreenⅠ.Results:To compare of 58 cases of patients with RA and 29 cases of healthy people,TGF-βRⅡ,Smad4 gene express and Smad4/Smad7 in PBMC of RA patient were significantly higher(P<0.01),but Smad7mRNA expression was near to healthy people and there was no significant difference.TGF-βRⅡ,Smad4 gene express and Smad4/Smad7 in active RA patient and before treatment were respectively higher than inactive and after treatment RA patient,and were increased markedly inⅢfunction stage(P<0.01),but no obvious differences inⅠandⅡstage (P>0.05).TGF-βRⅡ,Smad4,Smad7 mRNA expression had no significant difference in thesyndrome types of TCM,and the ratio of Smad4 and Smad7 in heat -dampness group and phlegm-stasis group had significant difference with cold group.There were positive correlations among Smad4 and TGF-βRⅡ,the ratio of Smad4 and Smad7(r1=0.262,P<0.05,r2=0.268,P<0.05),the level peak of TGF-βRⅡ,Smad4 were in 5-10 years.Conclusions:TGF-βRⅡ,Smad4 gene express and Smad4/Smad7 in PBMC of RA patient were significantly higher;in active RA patient and before treatment were respectively higher than inactive and after treatment RA patient;and were increased more inⅢfunction stage(P<0.01) than inⅠandⅡstage;and the ratio of Smad4 and Smad7 in heat -dampness group and phlegm-stasis group had significant difference with cold group.It may be an important observation factor of diagnosis,activity and prognosis and the destruction of the joint in RA to detect expression of the genes associated with TGF-β/Smad signaling pathways in RA patient.
