Objective To discuss the relationship between homocysteine (HCY) and congenital heart defects ,and to observe toxic effect of different dosage homocysteine on embryonic heart development in mammalian.Methods 30 S-D pregnant rats were randomly divided into three groups: the high dosage group(10), the low dosage group(lO) and the control group(lO). The first two groups were injected intraperitoneally with HCY 200mg/(kg·d) and 100mg/ (kg·d) respectively,while the control group was given the same volume physiologic saline intraperitoneally from 7 days pregnancy to 20 days pregnancy of uterine-incision delivery. The amounts,the weight,the length,the fetal death of juvenile rats were measured. The contents of HCY in serum were analyzed by high performance liquid chromatogram electrochem before pregnancy and 20 days pregnancy. The changes of structure of juvenile rats heart were observed by means of stereoscope. The changes of ultrastructure of juvenile rats cadiocyte were observed through transmission electron microscope.Results1 . HCY in serum of 20 days pregnant rats in the high dosage group was significantly higher than that in the control (30.47±1.12umol/L vs 10.98±0.77 umol/L, P=0.000, P<0.001), HCY in serum of 20 days pregnant rats in the low dosage group was significantly higher than that in the control (20.90±1.08umol/L vs 10.98±0.77 umol/L, P=0.000,P<0.001), and the hyperhomocysteinemia animal model was done well.2 . There were statistical significance in the incidence rate congenitalheart defects of juvenile rats between high dosage group and the control(14.13% vs 0.76%, p=0.000,p<0.001) .There were statistical significancein the incidence rate congenital heart defects of juvenile rats between lowdosage group and the control. 9.57% vs 0.76%, p=0.001, p<0.01)3. There were statistical significance in the quantity,the length and the rate of death of juvenile rats between low dosage group and the control (p<0.05) .There were statistical significance in the weight of juvenile rats between low dosage group and the control (p<0.001) .There were statistical significance in the quantity,the length and the weight of juvenile rats between high dosage group and the control (p<0.001) .There were statistical significance the rate of death of juvenile rats between high dosage group and the control (p<0.01) .4. The apoptosis of myocardial cell was seen in the low dosage group, and the apoptosis of myocardial cell was increased significantly in the high dosage group.Conclusions1. Hyperhomocysteinemia may be toxic effect on embryonic development of pregnancy rats and mainly expressing decreas quantity of juvenile rats, juvenile rats growth ristriction, malformation and fetal death.2. Hyperhomocysteinemia induced the apoptosis of myocardial cell, it may be one of the mechanisms that lead to congenital heart defects.
