Objective:To study the effect of volatile oils from Boswellia carterii Birdwood (VBCB),its liposome and 3-Keto-tirucall-8,24-dien-21-oic acid(KTDA)on the growth and proliferation on human MCF-7,SMMC-7721,K562,Hela and Tca 8113,and their the possible anticancer mechanisms.Methods:Effects of proliferation inhibition were detected on human MCF-7, SMMC-7721,K562,Hela and Tca 8113 treated with VBCB,its liposome and KTDA respectively with MTT assay;The morphologic changes of five cancer cells were observed by acirdine staining;and DNA fragmentation of the five ones was detected by agarose gel electrophoresis.Inhibitory rate and the change of cell-cycle were assessed by VBCB with Flow Cytometry(FCM);The change of expression of protein Bax and Bcl-2 were observed by VBCB with indirect immunofluorescence microscope.Results:1.VBCB could inhibit the growth of cancer cells at the concentration of 700,70,7,0.7,0.07 ug.mL-1for 24h,which depended on the concentration.At the concentration of 70 ug.mL-1treated for 48h or 72h,the "DNA ladder" was clearly observed by agarose gel electrophoresis.Apoptotic peak was exhibited by FCM(72h),and were oppressed at G1 phase on SMMC-7721 cell. Importantly,up-regulating expression of Bax was detected,the expression of Bcl-2 have no obvious change.2.VBCB(at 700,70,7 ug.mL-1)and its liposome(at 143,14.3,1.43 ug.mL-1)could inhibit five cancer cells growth treated for 72 h,which depended on the concentration.At the concentration of VBCB(70 ug.mL-1)and its liposome(14.3 ug.mL-1)for 72 h,apoptosis fragmentation was watched by fluorescence microscope,and the "DNA ladder" was clearly observed in agarose gel electrophoresis.3.KTDA(at 100,10,1 ug.mL-1)could inhibit the five cells growth treated for 72 h, which depended on the concentration.With 10 ug.mL-1KTDA treated for 72 h, apoptosis fragmentation was detected by fluorescence microscope,and the "DNA ladder" was clearly observed in agarose gel electrophoresis.Conclusion:1.VBCB,its liposome and KTDA could inhibit the reproduction of the SMMC7721 cells,inhibitory rate showed the dependence of concentration and time(only VBCB).2.VBCB could inhibit the reproduction of SMMC - 7721 cells,and might induce apoptosis of SMMC- 7721 cells through up-regulating the expression of Bax/Bcl-2,while the cells were oppressed at G1 phase.3.Base on IC50,VBCB and its liposome,KTDA were sensitive firstly on MCF-7, SMMC - 7721,K562,Hela and Tca 8113 secondly,and the drugs might lead to the inhibitory growth of the five ones through induced apoptosis.
