Objectives To explore the expression of transforming growth factor-beta1(TGF-β1)and connective tissue growth factor(CTGF)in children with congenital heart disease(CHD)and analysis the relationship of TGF-β1 and CTGF with the type of CHD(acyanotic congenital heart disease and cyanotic congenital heart disease),age,and the oxygen saturation.To investigate roles of TGF-β1 and CTGF in the development of myocardial fibrosis in CHD.Methods Of 45 childeren having CHD,25 were acyanotic congenital heart disease and 20 were cyanotic congenital heart disease.A small amount of fresh myocardium was cut from the right atrial appendage.To explore the expression of typeⅠ,typeⅢcollagen,TGF-β1 and CTGFmRNA through reverse transcription- polymerase chain reaction(RT-PCR).SPSS 13.0 software was used for the statistical analysis of these data.Results(1)The myocardial fibrosis of cyanotic congenital heart disease was more significant than acyanotic congenital heart disease (typeⅠcollagen p<0.01;typeⅢcollagen p<0.01).(2)The expression of TGF-β1 and CTGF mRNA were higher in cyanotic congenital heart disease than acyanotic congenital heart disease(p<0.001).(3)The expression of typeⅠ,typeⅢcollagen mRNA were correlated positively with age(typeⅠcollagen r=0.331,p<0.05;typeⅢcollagen r=0.372,p<0.05)and were negatively with oxygen saturation(typeⅠcollagen r=-0.561,p<0.001;typeⅢcollagen r=-0.537,p<0.001).(4)The expression of TGF-β1 and CTGF mRNA were correlated positively with the level of typeⅠ(TGF-β1 r=0.325,p<0.05;CTGF r=0.559,p<0.001), typeⅢcollagen mRNA(TGF-β1 r=0.403,p<0.05;CTGF r=0.331, p<0.05)and the degree of defection(TGF-β1 r=0.416,p<0.05;CTGF r= 0.563,p<0.001).They were negatively with oxygen saturation(TGF-β1 r=-0.631,p<0.001;CTGF r=-0.744,p<0.001).(5)The expression of CTGF mRNA in congenital heart disease was correlated positively with the expression of TGF-β1 mRNA(r=0.616,p<0.001).Conclusions(1)The myocardial fibrosis of cyanotic congenital heart disease was more significant than acyanotic congenital heart disease.(2) The expression of CTGF and TGF-β1 in cyanotic congenital heart disease were higher than acyanotic congenital heart disease.TGF-β1 and CTGF not only take part in but also play an important role in myocardial fibrosis with CHD especially C-CHD.The expression of CTGF mRNA in congenital heart disease was correlated positively with the expression of TGF-β1 mRNA.(3)The myocardial fibrosis in CHD was correlated with age and hypoix.(4)The expression of CTGF and TGF-β1 were negatively with oxygen saturation and correlated positively with the degree of defection.It is suggested that hypoix and load capacity can induce the expression of TGF-β1 and CTGF.
