Objectives To explore the expression of osteopontin in children with congenital heart disease (CHD) and analysis the relationship of OPN with the type of CHD (acyanotic congenital heart disease and cyanotic congenital heart disease), the oxygen saturation and age so as to investigate the roles of OPN in the development of myocardial fibrosis in CHD.Methods Of 45 childeren having CHD,25 were acyanotic congenital heart disease and 20 were cyanotic congenital heart disease. A small amount of fresh myocardium was cut from the right atrial appendage. To explore the expression of typeⅠ, typeⅢcollagen, OPNmRNA through reverse transcription-polymerase chain reaction(RT-PCR). SPSS 13.0 software was used for the statistical analysis of these data.Results (1) The expression of typeⅠ, typeⅢcollagen mRNA in cyanotic congenital heart disease was more significant than acyanotic congenital heart disease (typeⅠcollagen p<0.01; typeⅢcollagen p<0.01). (2) The expression of OPN mRNA was higher in cyanotic congenital heart disease than acyanotic congenital heart disease(p<0.05). (3) The expression of typeⅠ, typeⅢcollagen mRNA were correlated positively with age (typeⅠcollagen r=0.331, p<0.05; typeⅢcollagen r=0.372, p<0.05) and were negatively with oxygen saturation (typeⅠcollagen r=-0.561, p<0.001; typeⅢcollagen r=-0.537, p<0.001). (4) The expression of OPN mRNA was correlated positively with the level of type I (OPN r=0.625, p<0.001), typeⅢcollagen mRNA(OPN r=0.287, p<0.05) and the degree of defection(OPN r= 0.524, p< 0.001). It was negatively with oxygen saturation (OPN r=-0.703, p<0.001). Conclusions (1) The expression of typeⅠ, typeⅢcollagen mRNA in cyanotic congenital heart disease was more significant than acyanotic congenital heart disease. (2) The expression of OPN mRNA was higher in cyanotic congenital heart disease than acyanotic congenital heart disease. OPN not only takes part in but also plays an important role in myocardial fibrosis with CHD especially C-CHD. (3) The myocardial fibrosis in CHD was correlated with age and hypoix. (4) The expression of OPN was negatively with oxygen saturation and correlated positively with the degree of defection. It is suggested that hypoix and load capacity can induce the expression of OPN.
