Objective : To observe the effect of Buyanghuanwu Decoction (BHD) on hemorheology and ntric oxide (NO) of the high fat rats, it is to estimate objectively the function on regulating blood lipid and preventing athersclerosis (AS), via animal experiment, at the same time, investigate their functional mechanism. It can provide the experiment basis for using BHD to prevent and treat hyperlipidemia (HLP) as well as relative diseas in clinic.Methods: Sixty SD rats were randomly divided into six groups, each group consisted of ten rats. The six groups were as follows: blank control group, model group, low-dosage BHD group, medium-dosage group, high-dosage group and positive medicine (Xuezhikang) group. All the group were fed with high-fat feeder (total cholesterol 2%, sodium 0.2%, propyl sulf-oxygen pyridine 0.2%, lard 10%, normal feeder 87.6%), except the normal contrast group, which were fed with normal feeder. At the same time, the normal contrast group and the model contrast group were given distilled water, low-dosage BHD group were given 14g dried medicinal herb/kg, middle-dosage group were given 21g dried medicinal herb/kg, high-dosage group were given 28g dried medicinal herb/kg, positive medicine (Xuezhikang) group were given 0.12g xuezhikang /kg. The experiment was continued for eight weeks. After the last drug's given, absolute diet for 12 hours the rats were beheaded, blood was obtained. The total cholesterol(TC), triglyceride(TG), high density lipoprotein(HDL-C), low density lipoprotein(LDL-C), hematocritindex(HCT), fibrinogen(FIB), erythrocyte dimentation rate(ESR), ntric oxide(NO), were detected.Results:1 The effect on TC, TG, HDL-C, LDL-CContents TC (11.78±1.88, unit mmol/L, the following as the same), TG (2.40±0.44, unit mmol/L, the following as the same), LDL-C (3.12±0.38,unit mmol/L, the following as the same) in serum of model group, were significantly highter than those of normal group; HDL-C(0.31±0.08, unit mmol/L, the following as the same) were significantly lower than those of normal group, TC (H: 8.48±1.28;M: 8.10±0.69; L: 8.60±0.84), TG(H: 1.01±0.26; M: 0.85±0.14; L: 1.12±0.27;), LDL-C(H: 1.77±0.34; M: 1.65±0.35; L: 1.98±0.34)of BHD groups were significantly lower than those of model group ( p<0.01); HDL-C(H: 0.55±0.12; M: 0.54±0.14; L: 0.54±0.20) were significantly higher than those of model group, which suggested that BHD had obvious regulatory function on fat metabolism disturbance of rats on high-fat diet. Compared to Xuezhikang group, all of BHD groups were superior to Xuezhikang in creasing HDL-C, but there was no significant difference between the others indexes of BHD groups and Xuezhikang group (p>0.05).2 The effect on whole blood viscosity, hematocrit index(HCT), fibrinogen(FIB), erythrocyte sedimentationrate(ESR)Compared to normal group, whole blood viscosity (H: 4.73±0.37; M: 5.61±0.05; L 13.91±0.05 unit mps, the following as the same), HCT( 52.50±1.43 unit %, the following at the same), ESR (9.99±0.18 unitmm/h, the following as the same), FIB( 6.24±0.16 unitg/l, the following as the same) of model group significantly increased (p<0.01); Compared to model group, whole blood viscosity, hematocrit HCT, erythrocyte ESR, FIB of BHD groups significantly reduced (p<0.01), which suggested that BHD had obvious regulatory hemorheology of rats on high-fat diet. Compared to Xuezhikang group, BHD medium-dosage group and high-dosage group groups were superior to Xuezhikang in adjusting FIB.2 The effect on NOCompared to normal group, NO (12.34±1.31, unitμmol/ml, the following as the same) of model group significantly reduced (p<0.01); Compared to model group, NO (L: 16.62±1.01; M: 17.84±0.89; H: 19.8±0.70) of BHD groups significantly increased (p<0.01). There was no significantly difference between NO contents of BHD middle-dosage, low-dosage groups and Xuezhikang group (p>0.05).Conclusion:1 High-fat diet can disturb fat metabolism of rats. BHD had obvious regulatory function on fat metabolism disturbance of rats on high-fat diet.2 BHD can lower markedly whole blood viscosity. Their principle of reducing whole blood viscosity is reducing HCT, decreasing FIB and ESR. It also can restrain the abnormal rising of blood-lipid.3 BHD can increase NO in serum. It indicated that BHD could protect and improve vascular endothelial cell's function let the vascular endothelial cell play it's normal role of adjusting vessel's tension, protesting blood clotting and monocyte's adhersion, so to maintain the blood's flowing stability and thus prevent the emerging and developing of AS. So BHD could prevent and treat hyperlipidemia (HLP) as well as relative diseas in clinic.
