Object: To establish the rat model of infrarenal abdominal aorta occlusion, the contents of ET-1, SOD, and MDA in plasma and in renal homogenate were measured in each group, and we observed pathological changes in kidney and muscles of lower limb at different times. To study the effect of ET-1, SOD and MDA on kidney reperfusion injury caused by infrarenal abdominal aorta occlusion in rat model and its possible mechanism.Methods: Forty-two healthy Wister rats were randomly divided into 6 groups as following ( n = 7 ) : the control group; simply ischemia 5h without reperfusion(group I) ; 2 hour reperfusion following ischemia 5h (I/R2h) , 4 hours reperfusion following ischemia 5h (I/R4h) , 8 hours reperfusion following ischemia 5h (I/R8h) and 12 hours reperfusion following ischemia 5h (I/R12h). In each group the rats were killed to obtain samples of blood and kidney at the specified time points. The contents of ET-1, MDA, SOD in plasma and in renal homogenate were measured in each group. We observe the morphological changes of kidney and muscles of lower limb by light microscope and electron microscope.Results: ET-1 level of plasma in model I/R2h , I/R4h , I/R8h groups were higher obviously than these of control group, ET-1 level of plasma in model I/R4h group were higher obviously than these of group I ; ET-1 level of renal homogenate in model group I , I/R2h , I/R4h , I/R8h , I/R12h groups were higher obviously than these of control group, which were maximal in I/ R4h group , then decreased . SOD level of plasma in model I/R4h, I/R8h groups were lower obviously than these of control group, SOD level of plasma in model I/R4h, I/R8h groups were lower obviously than these of group I ; SOD level of renal homogenate in model group I, I/R2h, I/R8h, I/R12h groups were higher obviously than these of group I/R4h , which were minimal in I/R4h group , then increased. MDA level of plasma in model group I, I/R2h , I/R4h , I/R8h , I/R12h groups were higher obviously than these of control group, which were maximal in I/R4h group, then decreased. By light microscope and electron microscope: The injury degrees of kidney and muscles of lower limb in ischemia group were slight, the injury degree of reperfusion group was more severe than ischemia group.Conclusion: The kidney reperfusion injury caused by infrarenal abdominal aorta occlusion in rat model, it might be concerned with the increase of ET-1, lipid peroxidation damage after ischemia-reperfusion injury of lower limbs.
